摘 要:为进一步提高尼氏染色技术在厚切片形态学分析中的应用价值,以长爪沙鼠(Meriones unguiculatus)200μm冰冻脑切片为研究对象,在薄切片尼氏染色方法的基础上,对烘片、固定、洗涤和染色等实验步骤加以改良.结果发现,利用改良后的方法所制得的切片着色良好,脑区边界清晰,组织结构明显,无冰晶空洞和脱片现象.该实验结果可为科研工作者应用厚切片开展神经组织形态学研究提供重要参考.To improve the utility of the Nissl staining technique for morphological analysis of thick sections,200μm frozen brain sections from Meriones unguiculatus were used as the research object.Adapting the Nissl staining method traditionally used for thin sections,this study refined the procedure through steps including baking,fixing,washing and dyeing.The results demonstrated that the modified method yield sections with robust obtaining,devoid of cavities from ice crystals or detachment issues.The boundaries of the brain regions were distinct and the expression of the organizational structure was prominent.This experimental approach provides valuable insights for researchers aiming to apply thick sections in neuroscience studies.
