摘 要:探索黄柏、连翘、五倍子、白头翁和蒲公英5种中药水提物对大肠杆菌生物被膜形成的影响。采用试管二倍稀释法测定5种中药水提物对大肠杆菌的最低抑菌浓度(MIC),通过银染法观察5种中药水提物对大肠杆菌生物被膜黏附作用的影响,采用结晶紫染色法检测5种中药水提物对大肠杆菌生物被膜的抑制作用。结果显示,黄柏、连翘、五倍子、白头翁和蒲公英等水提物对大肠杆菌的MIC分别为500 mg/mL、500 mg/mL、250 mg/mL、250 mg/mL和125 mg/mL。通过镜像图可观察到对照组中有大面积的黑斑,而5种用药组的黑斑面积明显减小。对照组的大肠杆菌生物被膜洗脱液OD值为0.555,黄柏、连翘、五倍子、白头翁和蒲公英等用药组的生物被膜洗脱液OD值分别为0.245、0.236、0.237、0.213和0.229,与对照组比较差异极显著(P<0.01),5种中药水提物对大肠杆菌生物被膜的抑制率分别为55.9%、57.3%、57.5%、61.6%和58.7%。结果表明,5种中药水提物均能有效抑制大肠杆菌生物被膜的形成。To explore the effects of five Chinese herbal water extracts of Cortex Phellodendri,Fructus Forsythiae,Galla Chinensis,Radix Pulsatillae and Herba Taraxaci on the biofilm formation of Escherichia coli.The MIC of five Chinese herbal water extracts on Escherichia coli were determined by tube double dilution method,the effects of five Chinese herbal water extracts on the adhesion of Escherichia coli biofilm were observed by silver staining method,and the crystal violet staining method was used to detect the inhibitory effects of five Chinese herbal water extracts on the biofilm of Escherichia coli.The results showed that the MIC values of the water extracts of Cortex Phellodendri,Fructus Forsythiae,Galla Chinensis,Radix Pulsatillae and Herba Taraxaci were 500 mg/mL,500 mg/mL,250 mg/mL,250 mg/mL and 125 mg/mL against Escherichia coli,respectively.It could be observed in the mirror image that a large number of areas of dark spots were visible in the control group,while the area of dark spots were significantly reduced in the five medication groups.The OD value of Escherichia coli biofilm eluate in the control group was 0.555,while the OD values of eluate in the medication groups of Cortex Phellodendri,Fructus Forsythiae,Galla Chinensis,Radix Pulsatillae and Herba Taraxaci were 0.245,0.236,0.237,0.213 and 0.229,respectively,which were highly significant(P<0.01)compared with the control group,and the inhibition rates of the five Chinese herbal water extracts on Escherichia coli biofilm were 55.9%,57.3%,57.5%,61.6%and 58.7%,respectively.Thus,all the five Chinese herbal water extracts were effective in inhibiting the formation of Escherichia coli biofilm.
