摘  要:急性髓系白血病(acute myeloid leukemia,AML)属于造血系统克隆性恶性疾病。为了寻求AML临床治疗的潜在靶点,预测急性髓系白血病相关转录因子C/EBPα在急性髓系白血病中的发病机制,利用基因表达数据库(gene expression omnibus,GEO)筛选出敲除Cebpa基因的实验组与正常造血干细胞间的差异表达基因,并对其进行基因本体(GO)功能分析、京都基因与基因组百科全书(KEGG)通路分析。结果显示:在86个差异表达基因中,共有44个上调基因和42个下调基因;差异基因具有参与RNA聚合酶Ⅱ启动子转录的正调控、造血、基因表达的正向调控等功能,并且参与JAK-STAT信号通路、MAPK信号通路、PI3K-Akt信号通路的传导;而C/EBPα也与信号通路中的蛋白具有互作关系。由此预测异常表达的C/EBPα可能通过影响信号通路来干预急性髓系白血病的发病与预后。Acute myeloid leukemia(AML)is a clonal malignant disease of the hematopoietic system.In order to investigate underlying targets for the clinical treatment of acute myeloid leukemia,we predicted the pathogenesis of transcription factor C/EBPαassociated with acute myeloid leukemia in acute myeloid leukemia.We used the gene expression omnibus to screen the differentially expressed genes between the experimental group that knocked out the Cebpa gene and normal hematopoietic stem cells,and analyzed the gene ontology(GO)function and kyoto encyclopedia of genes and genomes(KEGG)pathway.The results showed that among the 86 differen‐tially expressed genes,there were a total of 44 up-regulated genes and 42 down-regulated genes.Differential genes are mainly involved in the positive regulation,hematopoiesis,and gene expression positive regulation of the RNA polymerase II promoter transcription,and participate in the conduction of JAK-STAT signaling path‐way,MAPK signaling pathway,and PI3K-Akt signaling pathway.C/EBPαalso interacts with proteins in the sig‐naling pathway.So we predict that aberrant expression of C/EBPαmay affect the pathogenesis and prognosis of acute myeloid leukemia through the signaling pathway.
