姜黄素通过细胞信号通路治疗胃癌的研究进展

摘　　要：胃癌是常见的消化系恶性肿瘤之一,生存率较低,预后较差,其发生发展与多个信号通路异常调节有关,包括Wnt/β联蛋白通路、Janus激酶2/信号转导及转录活化因子3通路、磷脂酰肌醇-3-激酶/蛋白激酶B/哺乳动物雷帕霉素靶蛋白通路、核因子κB通路、Fas/胱天蛋白酶3通路与miR-21/人第10号染色体缺失的磷酸酶及张力蛋白同源基因通路,这些信号通路的异常表达能促进肿瘤细胞生长增殖,抑制细胞凋亡,还能影响血管生成及转移,加重瘤体进展。姜黄素是一种中药活性提取物,具有抗炎和抗肿瘤等药理作用,对胃癌有一定的治疗作用,并可通过靶向调控相关通路阻断胃上皮细胞癌变,达到防治胃癌的目的,但尚未在临床普遍应用,未来姜黄素对人体胃癌的作用机制仍需更深入研究。Gastric cancer is one of the common malignant tumors in the digestive system,with a low survival rate and poor prognosis.Its occurrence and development are related to abnormal regulation of multiple signaling pathways,including Wnt/β-catenin pathway,Janus kinase 2/signal transducer and activator of transcription 3 pathway,phosphatidylinositol-3-kinase/protein kinase B/mammalian target of rapamycin pathway,nuclear factor-κB pathway,Fas/caspase-3 pathway,and miR-21/phosphatase and tensin homologue deleted on chromosome ten gene pathway.Abnormal expression of these signaling pathways can promote tumor cell growth and proliferation,inhibit cell apoptosis,and also affect angiogenesis and metastasis,aggravating tumor progression.Curcumin is an active extract of traditional Chinese medicine with anti-inflammatory and anti-tumor pharmacological effects,which has certain therapeutic effects on gastric cancer and can block gastric epithelial cell carcinogenesis through targeted regulation of related pathways,achieving the goal of preventing and treating gastric cancer.However,it has not yet been widely used in clinical practice,and the mechanism of action of curcumin on human gastric cancer still needs further research in the future.
