摘  要:目的探讨中枢性阻断交感神经药物盐酸可乐定对昼夜节律紊乱诱导的大鼠肠道和心脏组织结构及肠道炎症改变的影响。方法将15只大鼠随机分为正常光照组、光照组和给药组,每组5只。正常光照组予正常光照(光照:8:00~20:00,黑暗:20:00~8:00),每日8:00予2 ml纯净水灌胃;光照组予24 h持续光照,每日8:00予2 ml纯净水灌胃;给药组予24 h持续光照,每日8:00予2 ml盐酸可乐定(10μg/kg)灌胃,持续12周。造模结束后,对大鼠进行10%水合氯醛腹腔注射麻醉,取血后处死,将分离的大鼠肠道和心脏组织放入4%多聚甲醛固定保存,观察经苏木精-伊红染色大鼠肠道和心脏组织的病理组织学改变,采用免疫组织化学法观察大鼠肠道中促炎性细胞因子白细胞介素(IL)-6、IL-10的表达,比较三组大鼠肠道中促炎性细胞因子IL-6、IL-10的阳性面积占比。结果光镜下,正常光照组大鼠肠道组织病理结果显示,黏膜层、黏膜下层、肌层及浆膜层结构较完整;黏膜表面被覆单层柱状上皮,柱状上皮细胞形态未见明显变性、坏死或脱落;肠道固有层内的大肠腺排列较为密集,杯状细胞数量正常,固有层内偶见散在分布的淋巴细胞或巨噬细胞。光照组大鼠肠道组织病理结果显示,黏膜层、黏膜下层、肌层及浆膜层结构较完整;黏膜表面被覆单层柱状上皮,柱状上皮细胞形态未见明显变性、坏死或脱落;肠道固有层内可见炎症细胞浸润,主要为中性粒细胞。给药组大鼠肠道组织病理结果显示,黏膜层、黏膜下层、肌层及浆膜层结构较完整;柱状上皮细胞未见明显坏死或脱落;肠道固有层内未见炎症细胞浸润。光镜下,正常光照组、光照组、给药组大鼠的心脏组织心外膜组织结构均完整,未见明显增生或炎性浸润;心肌膜心肌纤维呈螺旋状排列,未见肥大、萎缩变细或断裂;心肌纤维形态未见异常,胞质染色均匀,胞核居中,多呈长椭Objective To explore the effects of central sympathetic blockade drug clonidine hydrochloride on changes in intestinal and cardiac tissue structure and intestinal inflammation induced by circadian rhythm disorders in rats.Methods Randomly divide 15 rats into normal light group,light group,and medication group,with 5 rats in each group.The normal light group was given normal light(light:8:00-20:00,dark:20:00-8:00),and 2 ml of pure water was gavaged daily at 8:00;The light group was continuously illuminated for 24 hours,and 2 ml of pure water was administered orally at 8:00 am every day;The administration group was given 24 hours of continuous light and 2 ml of clonidine(10μg/kg)by gavage for 12 weeks.After the completion of modeling,the rats were anesthetized by intraperitoneal injection of 10%chloral hydrate.Blood was collected and euthanized.The isolated intestinal and cardiac tissues of the rats were fixed and stored in 4%paraformaldehyde.The pathological changes in the intestinal of the rats stained with hematoxylin eosin were observed,and the expression of pro-inflammatory cytokines interleukin(IL)-6 and IL-10 in the intestinal and cardiac tissues of the rats was observed using immune histochemical method,compare the proportion of positive areas of pro-inflammatory cytokines IL-6 and IL-10 in the intestinal of three groups of rats.Results Under the light microscope,the pathological results of the intestinal tissue of rats in the normal light group showed that the structures of the mucosal layer,submucosal layer,muscular layer,and serosal layer were relatively complete;The mucosal surface is covered with a single layer of columnar epithelium,and the morphology of columnar epithelial cells does not show significant degeneration,necrosis,or detachment;The large intestine glands in the lamina propria of the intestine are densely arranged,and the number of goblet cells is normal.Scattered lymphocytes or macrophages are occasionally seen in the lamina propria.The pathological results of the intestinal tissue of ra
