基于CSE/H_2S体系探讨葱白提取物对H_2S催化酶及趋化因子CX3CL1的作用机制

摘  要:目的研究葱白提取物对动脉粥样硬化(AS)大鼠CX3C趋化因子配体1(CX3CL1)、CX3C趋化因子受体1(CX3CR1)、胱硫醚β合成酶(CBS)、3-巯基丙酮酸硫基转移酶(3MST)表达的影响及相互作用机制。方法纯种健康雄性SD大鼠,SPF级,体重(200±20)g,共40只,随机选择10只作为正常组,另外30只制备大鼠AS模型,再随机分为模型组、葱白提取物组、辛伐他汀组,每组各10只。正常组以基础饲料喂养,模型组、葱白提取物组、辛伐他汀组均以高脂维生素D3饲料喂养。从第5周开始,葱白提取物组大鼠给予配置好的葱白提取物溶液10 mL/kg体质量灌胃,辛伐他汀组大鼠给予辛伐他汀混悬液10 mL/kg体质量灌胃;正常组和模型组均给予等容积生理盐水10 mL/kg体质量灌胃,共连续8周。在第12周时,留取大鼠血液,分离主动脉。采用生化检测、HE染色、Western blot、RT-PCR检测脂质代谢、H_(2)S催化酶、趋化因子及基质金属蛋白酶表达。结果与模型组比较,葱白提取物组及辛伐他汀组大鼠血清总胆固醇、甘油三酯、低密度脂蛋白胆固醇、载脂蛋白B浓度均降低(P<0.05),高密度脂蛋白胆固醇浓度升高(P<0.05)。与模型组比较,葱白提取物组及辛伐他汀组斑块内部的胆固醇结晶较少,管腔狭窄程度较轻。与模型组比较,葱白提取物组和辛伐他汀组CX3CL1 mRNA、CX3CR1 mRNA、MMP7 mRNA、MMP9 mRNA表达均下调。与模型组比较,葱白提取物组和辛伐他汀组大鼠主动脉组织中3MST、CBS蛋白相对表达量升高(P<0.05),而CX3CL1、CX3CR1蛋白相对表达量降低(P<0.05)。结论葱白提取物可通过调控内源性H_(2)S生成体系,抑制CX3CL1、CX3CR1表达、降低MMP7、MMP9活性,发挥抗动脉粥样硬化作用。Objective To investigate the effects of scallion stalk on the expressions of CX3CL1,CX3CR1,CBS and 3MST in rats with atherosclerosis(AS)and its interaction mechanism.Methods A total of 40 purebred healthy male SD rats,SPF grade,weighing(200±20)g were fed in this research.10 rats were randomly selected as normal group,and the other 30 rats were prepared AS model.The rats were randomly divided into model group,scallion extract group and simvastatin group,10 rats in each group.Normal group was fed with basic diet,model group,scallion white extract group and simvastatin group were all fed with high fat vitamin D3 diet.Starting from the 5th week,the scallion white extract group was given 10 mL/kg body mass gavage,and the simvastatin group was given simvastatin suspension 10 mL/kg body mass gavage.Normal group and model group were given equal volume normal saline 10 mL/kg body massgavage for 8 weeks.At week 12,blood was taken from the rats and the aorta was isolated.Lipid metabolism,H_(2)S catalytic enzyme,chemokines and matrix metalloproteinases were detected by biochemical assay,HE staining,Western blot and RT-PCR.Results Compared with the model group,the concentrations of total cholesterol,triglycerides,low-density lipoprotein cholesterol,and apolipoprotein B in serum of scallion white extract group and simvastatin group were decreased(P<0.05),while the concentrations of high-density lipoprotein cholesterol were increased(P<0.05).Compared with the model group,the cholesterol crystals in the plaque ofscallion white extract group and simvastatin group were less,and the degree of lumen stenosis was less.Compared with the model group,the expression of CX3CL1 mRNA,CX3CR1 mRNA,MMP7 mRNA and MMP9 mRNA in the scallion white extract group and simvastatin group were down-regulated.Compared with the model group,the relative expression levels of 3MST and CBS proteins in the aortic tissues of the scallion white extract group and simvastatin group were increased(P<0.05),while the relative expression levels of CX3CL1 and CX3CR1 p
