摘  要:[目的]分析新月弯孢霉转录组,对甾体C_(11)β-羟基化酶基因进行鉴定。[方法]结合生物信息学和蛋白结构学对转录组分析,利用AlphaFold2进行蛋白结构模拟,与已知人源醛固酮合酶hCYP11B 2进行结构比对。[结果]在转录组中,共获得24943条Unigenes,平均长度1915 bp。18792条(75.34%)可以匹配到蛋白数据库获得注释信息。Unigenes在各数据库中注释的基因数分别为:在COG数据库中有5615条(22.51%),在GO数据库中有4424条(17.74%),在KEGG数据库中有6322条(25.35%),在Swiss-Prot数据库中有11348条(45.50%)。通过初筛获得39个基因。经过AlphaFold2的结构模拟,与人的甾体醛固酮合酶(hCYP11B2)结构比对进行复筛,共26条甾体C_(11)β-羟基化酶相关基因被鉴定出来。[结论]该新月弯孢霉菌株具有甾体C_(11)β-羟基化的能力,通过对其转录组进行分析,共鉴定出26条甾体C_(11)β-羟基化酶相关基因。[Objective]Analysis of the transcriptome of Curvularia crescent and identification of the steroid C_(11)β-hydroxylase gene.[Method]Combined bioinformatics and protein structure analysis of the transcriptome,protein structure simulation using AlphaFold2,and structure comparison with known human-derived aldosterone synthase hCYP11B2.[Result]In the transcriptome,a total of 24943 Unigenes were obtained with an average length of 1915 bp.18792(75.34%)could be matched to protein databases to obtain annotation information.the number of genes annotated by Unigenes in each database were:5615(22.51%)in the COG database,4424 in the GO database(17.74%),6322(25.35%)in the KEGG database and 11348(45.50%)in the Swiss-Prot database.Thirty-nine genes were obtained by primary screening.After structural simulation by AlphaFold2 and re-screening by structural comparison with human steroidal aldosterone synthase(hCYP11B2),a total of 26 steroidal C_(11)β-hydroxylase related genes were identified.[Conclusion]This strain of has the ability of steroid C_(11)β-hydroxylation,and a total of 26 steroid C_(11)β-hydroxylase related genes were identified by analysis of its transcriptome.
