摘 要:【背景】种子贮藏对保障种子生产具有重要意义。【方法】对烟草种子贮藏期间的转录组和蛋白组进行分析。【结果】贮藏后的烟草种子发芽率和发芽势逐渐提高并趋于稳定,贮藏后种子充分成熟。贮藏前后共获得了差异基因264个,差异多肽256个,差异基因及差异多肽均以下调为主;差异基因的GO和KEGG分析主要富集到脂肪酸代谢、激素信号、防御反应等过程。差异多肽所属蛋白的GO和KEGG分析主要富集到脂肪酸代谢、过氧化酶体、胚胎发育等过程。另外,从差异多肽所属蛋白中筛选获得2个目标多肽,其中Nitab4.5_0008018g0020是类2S富硫种子贮藏蛋白2(2S sulfur-rich seed storage protein 2-like),与种子成熟过程的营养积累有关,Nitab4.5_0002257g0090无功能注释。此外,从差异基因和差异多肽所属蛋白的交集中鉴定到1个共有蛋白,功能注释为枯草菌素样蛋白酶SBT1.3,具有丝氨酸型内肽酶活性。【结论】本研究从转录和翻译水平解析了烟草种子贮藏期间的内部生命活动变化规律,为种子贮藏调控分子机理研究提供了组学研究基础。Seed storage is of significance for ensuring seed production.The transcriptome and proteome of tobacco seeds were analyzed during the storage period.As the result shown,after storage,the germination rate and vigor of tobacco seeds gradually increased and stabilized,and the seeds were fully matured.A total of 264 differentially expressed genes and 256 differentially expressed peptides were obtained before and after storage,with a down-regulation predominance.The GO and KEGG enrichment analyses of differentially expressed genes were mainly enriched in processes such as fatty acid metabolism,hormone signaling,and defense responses.The GO and KEGG enrichment analyses of the proteins corresponding to differentially expressed peptides were mainly enriched in processes such as fatty acid metabolism,peroxisomes,and embryonic development.In addition,two target peptides were further screened from the proteins corresponding to the differentially expressed peptides Among them,Nitab4.5_0008018g0020 is a Class 2S sulfur-rich seed storage protein 2-like,related to nutrient accumulation during seed maturation,and Nitab4.5_0002257g0090 has no functional annotation.Furthermore,a common protein annotated as subtilisin-like protease SBT1.3 with serine-type endopeptidase activity was identified from the intersection of differentially expressed genes and proteins corresponding to differentially expressed peptides.This study deciphered the internal life activity changes of tobacco seeds during storage at the transcript and translation levels,providing an omics research foundation for the study of molecular mechanisms regulating seed storage.
