摘  要:目的探讨姜黄素(curcumin)对递增负荷大强度运动大鼠肾脏的作用。方法44只7w龄雄性SD大鼠随机分为安静对照(C,n=12)、递增负荷大强度运动致肾脏损伤模型(M,n=16)和姜黄素干预(CM,n=16)3组。C组不进行运动干预,其他组进行6w递增负荷跑台训练(起始:速度10m/min,时间10min,坡度10°;终止:速度40 m/min,时间180 min,坡度10°)。每次训练前1 h,CM组灌胃姜黄素溶液[200 mg/(kg·bw),5 ml/(kg·bw)],其他组灌胃等体积0.5%羧甲基纤维素钠溶液。末次训练结束24 h后处死大鼠,取血及肾脏。肾脏HE染色后观察组织形态,采用原位末端凋亡法检测肾脏细胞凋亡水平,酶联免疫吸附法、免疫组织化学法等方法检测相关生化指标,包括:血清肌酐(serum creatinine,Scr)、血尿素氮(blood urea nitrogen,BUN),肾脏肿瘤坏死因子-α(tumor necrosis factor-α,TNF-α)、白细胞介素-1β(interleukin-1β,IL-1β)、白细胞介素-18(interleukin-18,IL-18)及沉默信息调节因子1(sirtuin 1,SIRT1)、核因子-κB p65(nuclear factor-kappa B p65,NF-κB p65)、乙酰化核因子-κB p65(acetylated nuclear factor-kappa B p65,Ac-NF-κB p65)、B细胞淋巴瘤-2(B-cell lymphoma-2,Bcl-2)、Bcl-2相关X蛋白(Bcl-2 associated X protein,Bax)。结果与C组相比,M组肾脏组织形态发生病理性改变,Scr、BUN水平极显著升高(P<0.01),肾脏SIRT1、Bcl-2蛋白质表达水平及Bcl-2/Bax极显著降低(P<0.01),肾脏细胞凋亡、Bax、NF-κB p65、Ac-NF-κB p65蛋白质表达及炎症因子TNF-α、IL-1β、IL-18水平极显著升高(P<0.01);与M组相比,CM组肾脏组织形态病理性改变明显改善,Scr、BUN水平极显著降低(P<0.01),肾脏SIRT1、Bcl-2蛋白质表达水平及Bcl-2/Bax显著升高(P<0.05或P<0.01),肾脏细胞凋亡、Bax、NF-κBp65、Ac-NF-κBp65蛋白质表达及炎症因子TNF-α、IL-1β、IL-18水平显著降低(P<0.05或P<0.01)。结论姜黄素能够调控SIRT1/NF-κB信号通路相关蛋白质表达,缓解炎症反应和细胞凋亡,保护大鼠�Objective To explore the effects of curcumin on the kidney of rats with incremental load high-intensity exercise.Methods Forty-four 7-week-old male SD rats were randomly divided into control group(C,n=12),incremental load high-intensity exercise-induced kidney injury model group(M,n=16)and curcumin intervention group(CM,n=16).There was no exercise treatment in C group,while the other groups underwent 6-week incremental treadmill training(start:velocity was 10 m/min,training time was 10 min,slope was 10°;end:velocity was 40 m/min,training time was 180 min,slope was 10°).One hour before training,CM group was given curcumin solution(200 mg/(kg·bw),5 ml/(kg·bw)),and the other groups were given an equal volume of 0.5%carboxymethylcellulose sodium solution.The rats were sacrificed 24 hours after the last training,and blood and kidney were collected.The morphology of kidney was observed after HE staining.The level of renal apoptosis was detected by TdT mediated dUTP nick end labeling.The related biochemical indexes,including serum creatinine(Scr),blood urea nitrogen(BUN)and renal tumor necrosis factor-α(TNF-α),interleukin 1β(IL-1β),interleukin 18(IL-18),sirtuin 1(SIRT1),nuclear factor-kappa B p65(NF-κB p65),acetylated nuclear factor-kappa B p65(Ac-NF-κB p65),B-cell lymphoma-2(Bcl-2),Bcl-2 associated X protein(Bax),were detected by enzyme linked immunosorbent assay,immunohistochemistry or other methods.Results Compared with group C,pathological changes were noted in the kidney in group M.The level of Scr and BUN were increased significantly(P<0.01).The protein expression level of renal SIRT1,Bcl-2 and Bcl-2/Bax ratio were decreased significantly(P<0.01).The level of renal apoptosis,protein expression of Bax,NF-κB p65 and Ac-NF-κB p65,TNF-α,IL-1βand IL-18 were increased significantly(P<0.01).Compared with group M,renal pathological changes in group CM were improved significantly.The level of Scr and BUN were decreased significantly(P<0.01).The protein expression level of renal SIRT1,Bcl-2 and Bcl-2/Bax ratio
