摘 要:建立了高效液相色谱—示差折光检测器(HPLC-RID)测定海藻碘液中甘露醇含量的分析方法。通过对色谱柱、流动相等色谱条件优化,选用Agilent Polaris NH_(2)色谱柱(250 mm×4.6 mm,5μm)为分析柱,乙腈和水(体积比为70∶30)为流动相,流速1.0 mL/min,柱温30℃,示差折光检测器(RID)检测。结果表明,甘露醇在0.1~5.0 mg/mL的浓度范围内,线性关系良好,相关系数为0.9998;加标回收率为97.2%~98.8%,相对标准偏差(RSD)小于3.0%。该方法操作简单,准确度好,精密度高,适用于海藻碘液中甘露醇的快速测定。A method for determination of mannitol in seaweed iodine concentrate solution by high performance liquid chromatography with differential refractive index detector(HPLC-RID) was established.The separation effect of mannitol and other components in seaweed iodine concentrate solution were investigated by optimizing the chromatographic conditions of column and mobile phase.The samples were separated by Agilent Polaris NH_2(250×4.6 mm,5 μm)column,and detected by HPLC with RID detector.The mobile phase was a mixture of acetonitrile and water(the volume is 70 ∶30) at a flow rate of 1.0 mL/min,and the column temperature was 30 ℃.The calibration curves of mannitol showed good linearity in the range of 0.1~5.0 mg/mL with the correlation coefficients of 0.999 8.The spiked recoveries of mannitol in seaweed iodine concentrate solution ranged from 97.2% to 98.8% with relative standard deviation(RSD) less than 3.0%.The results indicated that the proposed method showed the advantages of simplicity,good accuracyand high precision,and was suitable for the rapid determination of mannitol in seaweed iodine concentrate solution.
