摘 要:目的观察单侧耳蜗毁损后1、2、4周甘氨酸受体α1亚单位(glycine receptor alpha1 subunit,GlyRα1)在大鼠耳蜗核的表达变化,为探讨单侧聋听觉中枢重组的分子机制提供参考。方法SD大鼠在出生后12 d,随机分为实验组(左侧耳蜗毁损)及对照组,术后1、2、4周取耳蜗核,切片后采用免疫组化方法观察GlyRα1的表达变化。结果GlyRα1在两组双侧耳蜗核中的表达在造模后2周出现升高,4周降低;实验组术后2周双侧表达差异有统计学意义(P<0.05),术后1、4周及对照组各时间段双侧无统计学差异(P>0.05)。对照组左侧与实验组术侧相比术后1、4周差异有统计学意义(P<0.05),术后2周差异无统计学意义(P>0.05);实验组及对照组右侧术后1、2、4周均有统计学差异(P<0.05)。结论单侧听觉剥夺导致大鼠双侧耳蜗核接收不对称的信息,观察到的耳蜗核GlyRα1在发育中的不对称改变可能为初级听觉中枢重组所必需。OBJECTIVE To observe the expression changes of glycine receptorα1 subunit(GlyRα1)in the cochlear nucleus of rats 1,2 and 4 weeks after unilateral cochlear lesion,so as to provide a reference for exploring the molecular mechanism of the reorganization of the hearing center of unilateral deafness.METHODS Sprague dawley(SD)rats were fed until 12 days after birth,and were randomly divided into experimental group(left cochlear lesion)and control group.One week,two weeks,and four weeks after operation,bilateral Cochlear nucleus were taken from the two groups of rats,and immunohistochemical method was used to observe the expression of GlyRα1 in the Cochlear nucleus.RESULTS In both groups,the expression of GlyRα1 in the cochlear nucleus of the operative side and the healthy side increased 2 weeks after the model establishment,and decreased 4 weeks after the model establishment;In the experimental group,there was a significant difference in the expression of GlyRα1 between two sides 2 weeks after operation(P<0.05),while there was no significant difference 1 week and 4 weeks after operation(P>0.05);In the control group,there was no significant difference in the expression of GlyRα1 between two sides at each time period(P>0.05).The difference between the operation side of the control group and the experimental group was statistically significant 1,4 weeks after operation(P<0.05),and there was no significant difference 2 weeks after operation(P>0.05).Between the experimental group and the control group on the right side,there was a significant difference(P<0.05)at 1 week,2 weeks,and 4 weeks after surgery.CONCLUSION Unilateral auditory deprivation causes bilateral Cochlear nucleus of rats to receive asymmetric information,and the observed asymmetric change of GlyRα1 in Cochlear nucleus during development might be necessary for the reorganization of the primary auditory center.
