摘 要:为探究暗前远红光(EOD-FR)下赤霉素(GA)和油菜素内酯(BR)对南瓜下胚轴伸长生长的调控机制及二者之间的作用关系,以‘日本雪松’南瓜为试验材料,以EOD-FR处理为手段,研究外源增施GA、BR以及GA合成抑制剂(PAC)、BR合成抑制剂(BRZ)对下胚轴生长、激素含量和基因相对表达量的影响,并探讨GA和BR在暗前远红光介导的下胚轴伸长过程中的互作关系和重要作用。结果表明:EOD-FR可以显著提高39.8%的下胚轴长度,提高内源GA和BR的含量以及GA和BR合成与信号转导基因的表达量;同时添加GA和BR的下胚轴长度要显著高于单独添加BR和GA,而且PAC和BRZ也具有叠加效应;BR可以缓解28.9%的PAC对下胚轴的抑制作用,GA也缓解了30.8%的BRZ对下胚轴的抑制作用;BRZ显著降低下胚轴内6.7%的GA含量,抑制GA的合成和信号转导相关基因的表达;PAC显著降低了下胚轴内20.9%的BR含量,也抑制了BR合成和信号转导相关基因的表达。综上,在EOD-FR介导的下胚轴伸长生长中,GA和BR相互影响,协同调控南瓜下胚轴长度。In order to explore the regulation mechanism of gibberellin(GA)and brassinosteroids(BR)on hypocotyl elongation and growth of pumpkin under EOD-FR and their interaction relationship,this study took‘Japanese cedar'pumpkin as the experimental material.EOD-FR treatment was used to investigate the effects of exogenous GA,BR and GA synthesis inhibitors(PAC)and BR synthesis inhibitors(BRZ)on hypocotyl growth,hormone content and gene relative expression.The interaction and important role of GA and BR in hypocotyl elongation mediated by EOD-FR were discussed.The results showed that:EOD-FR could significantly increase the length of hypocotyls by 39.8%,the content of endogenous GA and BR,and the expression of GA and BR synthesis and signal transduction genes.The length of hypocotyls supplemented with both GA and BR was significantly longer than that those supplied with BR and GA alone,and PAC and BRZ also had additive effect.BR alleviated 28.9%of PAC inhibition on hypocotyl,GA alleviated 30.8%of BRZ inhibition on hypocotyl.BRZ significantly reduced 6.7%of GA content in hypocotyl,inhibited GA synthesis and expression of signal transduction related genes.PAC significantly reduced 20.9%of BR content in hypocotyl and also inhibited the expression of genes related to BR synthesis and signal transduction.To sum up,in EOD-FR mediated hypocotyl elongation,GA and BR interacted and coordinated to regulate the hypocotyl length of pumpkin.
