摘  要:目的应用人源性ras转基因小鼠进行26周短期致癌试验前,需要做1个4周剂量探索试验,可选的实验动物有ras转基因小鼠CB6F1(+)、同窝阴性鼠CB6F1(-)以及具有相同遗传背景的C57小鼠,本研究旨在比较3种小鼠特定阶段的主要生物学特征。方法选用5~6周龄的3种实验小鼠各20只,雌雄各半,比较4周内体重、摄食量与食物利用率的变化,以及检测4周末血常规及血清生化指标等。结果与相应性别的CB6F1(+)小鼠相比,雌性CB6F1(-)小鼠的WBC、RBC、HGB、ALB、TP和CHOL较高,以上差异均有统计学意义(F=7.283、4.349、6.081、91.577、21.670和11.673,P<0.05或P<0.01),第3周的食物利用率较低,差异具有统计学意义(F=15.662,P<0.01);雄性CB6F1(-)小鼠HCT较高,具有统计学意义(F=2.861,P<0.05)而EO%与UREA较低,以上差异均有统计学意义(F=3.308、9.852,P<0.05);雌性C57小鼠的体重、摄食量、食物利用率与HCT、PLT、NEUT%、GLU、CREA较低,以上差异均有统计学意义(F=2.512、24.573、10.995、5.931、8.926、4.628、39.916和6.311,P<0.05或P<0.01),WBC、RBC、HGB、LYMPH%、AST、ALB、TP、UREA和CHOL较高,以上差异均有统计学意义(F=7.238、3.349、6.081、4.195、11.236、91.577、21.670、26.267和11.673,P<0.05或P<0.01);雄性C57小鼠的体重、摄食量、食物利用率与PLT、NEUT%、GLU较低,以上差异均有统计学意义(F=16.546、109.173、18.724、7.694、23.728和24.680,P<0.05或P<0.01),LYMPH%、MONO%、ALT、AST、ALB、TP和CHOL较高,以上差异均有统计学意义(F=6.500、6.767、5.689、23.288、74.435、28.203、6.853,P<0.05或P<0.01)。CB6F1(-)小鼠与CB6F1(+)小鼠的主要生物学特征差异较小,而C57小鼠则明显较大。结论CB6F1(-)小鼠更适宜作为26周短期致癌试验剂量探索的实验动物,而应用C57小鼠时需适当地考虑种属差异。Objective A 4-week dose range-finding study is required before the operation of 26-week short-term carcinogenesis test using the human ras transgenic mice,and three types of animals are selected to be used,including ras transgenic mice CB6F1(+),littermates negative mice CB6F1(-)and the same genetic background of C57 mice.The purpose of this study is to compare the main biological characteristics of the 3 types of mice at specific stages.Methods Twenty mice at 5-6 weeks old of every kind animal were selected,half male and female.The changes of body weight,food intake and food utilization rate were compared,and the blood routine and serum biochemical indexes were detected in four weeks.Results Compared with CB6F1(+)mice of the corresponding sex,female CB6F1(-)mice had higher WBC,RBC,HGB,ALB,TP,CHOL,and the differences were statistically significant(F=7.283,4.349,6.081,91.577,21.670,11.673,P<0.05 or P<0.01)and lower food utilization in the third week,which was statistically significant(F=15.662,P<0.01),while male CB6F1(-)mice had higher HCT,which was statistically significant(F=2.861,P<0.05)and lower EO%,UREA,the differences were statistically significant(F=3.308、9.852,P<0.05).The body weight,food intake food utilization,HCT,PLT,NEUT%,GLU and CREA in female C57 mice were lower than the female CB6F1(+),the differences were statistically significant(F=2.512,24.573,10.995,5.931,8.926,4.628,39.916,6.311,P<0.05 or P<0.01)and WBC,RBC,HGB,LYMPH%,AST,ALB,TP,UREA and CHOL were higher,and the differences were statistically significant(F=7.238,3.349,6.081,4.195,11.236,91.577,21.670,26.267,11.673,P<0.05 or P<0.01).The body weight,food intake and food utilization rate,PLT,NEUT%,GLU in male C57 mice were lower than the male CB6F1(+),the differences were statistically significant(F=16.546,109.173,18.724,7.694,23.728,24.680,P<0.05 or P<0.01)and LYMPH%,MONO%,ALT,AST,ALB,TP and CHOL were higher,and the differences were statistically significant(F=6.500,6.767,5.689,23.288,74.435,28.203,6.853,P<0.05 or P<0.01).The differences of main
