摘 要:支气管哮喘是儿童常见的慢性气道炎症性疾病,其发病机制与遗传及环境因素密切相关。多项临床流行病学调查发现血清类黏蛋白1样蛋白3(orosomucoid 1-like protein 3,ORMDL3)基因单核苷酸多态性变异与儿童哮喘的发生密切相关,但是目前ORMDL3参与儿童哮喘发生发展的机制尚不明确。近年来研究人员对ORMDL3生物学功能进行探究,发现ORMDL3可能通过内质网应激、鞘脂代谢反应参与支气管上皮细胞炎症、上皮细胞屏障受损和T细胞免疫失衡等过程,也可能介导了婴幼儿早期呼吸道病毒感染诱发喘息的机制。因此,该文对ORMDL3在儿童支气管哮喘发病机制中的作用进展进行综述。Bronchial asthma is a common chronic airway inflammatory disease in children,and its pathogenesis is closely related to genetic and environmental factors.A number of clinical epidemiological investigations have found that the single nucleotide polymorphisms variation of orosomucoid 1-like protein 3(ORMDL3)gene is closely related to the incidence of childhood asthma,but the mechanism of ORMDL3′s involvement in the occurrence and development of childhood asthma remains unclear.In recent years,researchers have explored the biological function of ORMDL3,and found that ORMDL3 may be involved in bronchial epithelial cell inflammation,epithelial cell barrier damage and T cell immune imbalance through endoplasmic reticulum stress and sphingoid metabolic response,and may also mediate the mechanism of early respiratory tract infection and possible wheezing in infants and young children.Therefore,this paper reviews the latest progress of ORMDL3′s involvement in the pathogenesis of bronchial asthma in children.
