摘 要:细菌依赖双组分系统(Two-component system,TCS)的转导机制来感知和适应外界环境变化,并产生适应性反应,从而应对不利环境对自身的影响,达到增殖和入侵宿主的目的。TCS还可调控细菌毒力因子和抗生素耐药基因的表达。与传统抗生素以特定蛋白为靶点不同,TCS抑制剂主要靶向组氨酸蛋白激酶或反应调节蛋白,因此,研究TCS的作用机制可为降低细菌耐药性和毒力、新型抗菌药物的研发提供新思路。文中综述了TCS的作用机制、TCS对细菌毒力和耐药性的调控,以及近年来TCS抑制剂的研究进展,以期为新型抗菌药物的研发和病原菌的防控提供参考。Bacteria rely on the transduction mechanism of the two-component system(TCS) to sense and adapt to changes in the external environment and produce adaptive response,so as to cope with the impact of adverse environment on themselves and achieve the purpose of proliferation and invasion of the host.TCS can also regulate the expression of bacterial virulence factors and antibiotic resistance genes.Different from traditional antibiotics that target specific proteins,TCS inhibitors mainly target histidine protein kinase or response regulatory proteins.Therefore,studying the mechanism of TCS can provide new ideas for reducing bacterial drug resistance and virulence and developing new antibacterial drugs.This article reviews the mechanism of TCS,the regulation of TCS on bacterial virulence and drug resistance,as well as the research progress of TCS inhibitors in recent years,in order to provide references for the development of new antibacterial drugs and the prevention and control of pathogens.
