摘  要:为了探讨温胆汤含药血清对代谢相关脂肪性肝病(MAFLD)细胞模型药物干预的作用及可能的分子机制,将C57BL/6J雄性小鼠随机分为正常对照组和温胆汤含药血清组,实验第14 w处死小鼠,采取下腔静脉取血的方式,收集全血,制备含药血清.利用MTT方法检测不同浓度的温胆汤含药血清对于HepG2细胞活力的影响.利用游离脂肪酸建立MAFLD HepG2细胞模型,细胞模型分为6组:正常对照组(简称空白组)、细胞模型组(简称模型组)温胆汤含药血清低剂量组(简称低剂量组)、温胆汤含药血清中剂量组(简称中剂量组)、温胆汤含药血清高剂量组(简称高剂量组)、吡咯列酮治疗组(简称吡咯列酮组).比较6组细胞甘油三酯含量,检测各组细胞的炎症反应水平、氧化损伤指标丙二醛(MDA)、超氧化物歧化酶(SOD)水平、脂质代谢重要基因、蛋白SREBP-1c、FAS、DGAT-2、SCD-1和ACO表达水平.MTT检测的结果表明,5、10、20、和50 mg/L的温胆汤含药血清对于HepG2细胞活力不会产生明显的抑制作用,但是,100 mg/L的温胆汤含药血清会产生明显的细胞毒性.模型组细胞与空白组细胞比较,细胞中形成大量脂滴,甘油三酯含量明显升高,证实了游离脂肪酸成功构建了MAFLD细胞模型;各药物干预治疗组细胞脂肪水平,甘油三酯含量、炎症反应水平、脂质过氧化水平、和脂质代谢重要基因和蛋白表达均较模型组细胞显著降低,高剂量组和吡咯列酮组降低最为显著,差异有统计学意义(P<0.05).本研究表明不同剂量温胆汤含药血清均能明显改善MAFLD细胞模型中脂质沉积,降低甘油三酯含量和炎症反应水平,可能与调节脂质代谢、降低细胞中脂质过氧化水平和脂质代谢重要基因的表达有关.The purpose of this study is to explore the intervention effect of Wendan decoction containing serum on metabolic-related fatty liver(MAFLD)cell models and its possible molecular mechanisms.C57BL/6J male mice were randomly divided into normal control and Wentan decoction containing serum groups.The mice were sacrificed at the end of the 14 w,and the whole blood was collected by the inferior vena cava to prepare drug-containing serum.The effect of diferent concentrations of drugcontaining serum of Wendan decoction on HepG2 cell viability was determined by the MTT method.MAFLD HepG2 cell model was established using free fatty acids.The cell model was divided into 6 groups.Normal control group(blank group),cell model group(model group)Wentan decoction medicated serum low dose group(low dose group),Wentan decoction medicated serum medium dose group(medium dose group),Wentan decoction medicated serum high dose group(high dose group),pyrrolilitone treatment group(pyrrolilitone group);The levels of fat deposition and triglyceride content of the 6 groups were compared,and the levels of the inflammatory response,oxidative damage indicators malondialdehyde(MDA),superoxide dismutase(SOD)and expression levels of important genes and proteins SREBP-lc,FAS,DGAT-2,SCD-1 and ACO in lipid metabolism were detected in each group.The results of MTT assay showed that 5,10,20 and 50 mg/L medicated serum of Wentan decoction did not have significant inhibitory effect on HepG2 cell viability,but 100 mg/L medicated serum of Wentan decoction could produce significant cytotoxicity.Compared with the cells in the blank group,a large number of lipid droplets were formed in the model group,and the content of triglyceride was significantly increased,which confirmed that the free fatty acids successfully constructed the MAFLD cell model.Compared with the model group,the levels of fat,triglyceride content,inflammatory response level,lipid peroxidation level and the expression of important genes and proteins in lipid metabolism were significantly de
