摘  要:目的 检测兔膀胱出口部分梗阻(partial bladder outlet obstruction,pBOO)及解除后膀胱功能和细胞超微结构的改变。方法 将24只雄性新西兰白兔分为梗阻组、梗阻解除组和对照组,每组8只。梗阻组动物行手术部分结扎膀胱颈部,建立pBOO动物模型。随后进行膀胱容量、膀胱重量测试,并提取兔逼尿肌组织,于透射电镜下对细胞内线粒体、内质网等超微结构进行观测,并于多种缓冲液中对逼尿肌束进行肌肉收缩力检测。结果 对照组膀胱重量明显低于其他两组,在除无Ca^(2+)液以外溶液中对照组单位重量逼尿肌最大收缩速度及最大张力都明显大于梗阻解除组,而梗阻组与梗阻解除组之间无明显差异。梗阻组较对照组内质网明显扩张,线粒体水肿明显,且相对密度下降。讨论梗阻2周后兔逼尿肌内质网与线粒体均发生形态学改变且未能恢复正常,此变化可能同逼尿肌膀胱功能下降相关。Objective To detect the changes of rabbits’bladder function and cell ultrastructure after partial bladder outlet obstruction(pBOO)with its subsequent relief.Methods Twenty-four male New Zealand white rabbits were divided into obstruction group,pBOO relief group and control group,with 8 rabbits in each group.The animals in the obstructive group were operated by partial ligating the bladder neck to form the pBOO animal model.Subsequently,bladder capacity and bladder weight were compared,and rabbit detrusor tissue was extracted.The ultrastructures such as mitochondria and endoplasmic reticulum in cells were observed under transmission electron microscope.Contractility test was processed in various buffers.Results The bladder weight of the control group was significantly lower than that of the other two groups.In solutions other than Ca^(2+)-free,the maximum contraction velocity and maximum tension of the detrusor per unit weight in the control group were significantly greater than those in the other two groups.No significant difference was detected between obstruction and pBOO relief groups.Compared with the control group,the endoplasmic reticulum in the obstructed group was significantly expanded,and the mitochondrial was obviously swollen,and the relative density decreased.Conclusion Detrusor endoplasmic reticulum and mitochondria of rabbits had morphological changes after 2 weeks of obstruction and failed to return to normal level.This change may be related to the decline of detrusor bladder function.
