摘 要:亚麻(Linum usitatissimum L.)是世界上重要的纤维作物,赤霉素对亚麻的生长发育起着重要的调控作用。通过对亚麻赤霉素氧化酶基因GA2ox、GA3ox和GA20ox进行全基因组鉴定和序列分析,并运用转录组和实时荧光定量PCR技术分析亚麻快速生长期茎尖中GAoxs的表达水平。本研究从亚麻全基因组共鉴定出23个赤霉素氧化酶基因,其中GA2ox类11个、GA3ox类4个,GA20ox类8个,分布于除6、7、9号染色体外其他染色体上;亚麻赤霉素氧化酶基因包含2~4个外显子;蛋白质理论分子量在35.36~47.71 kD之间;等电点为5.25~8.90;基因结构和保守结构域分析发现motif 1、motif 2、motif 3、motif 4、motif 5、motif 6、motif 9、motif 10是大部分基因所共有的,motif 8是GA2ox所特有的。系统发育分析将23个赤霉素氧化酶基因聚类在4个分支上。转录组结果表明,在高秆和矮秆亚麻快速生长期的茎尖中有6个基因差异表达,分别为4个GA2ox (LuGA2ox5,LuGA2ox6,LuGA2ox10,LuGA2ox11)和2个GA20ox (LuGA20ox5,LuGA20ox7)。结合各基因在亚麻组织中的特异性表达分析,表明在茎尖特异性表达的LuGA20ox7可能与营养生长期的株高形成相关。本研究为解析亚麻赤霉素氧化酶基因的结构特征及其在亚麻生长发育中的表达模式提供了理论参考。Flax(Linum usitatissimum L.) is one of the most important fiber crops in the world.Gibberellin plays an important role in regulating the growth and development of flax.Genome-wide identification and sequence analysis of GA2ox,GA3ox and GA20ox genes in flax were performed.Real-time PCR was used to analyze the expression level of GAoxs in the shoot tip for the rapid growth period.In this study,a total of 23 GAoxs were identified with the genome-wide of flax,including 11 GA2ox genes,4 GA3ox genes,and 8 GA20ox genes,which were common to other chromosomes except chromosome 6,7,and 9.The GAoxs of flax contain 2~4 exons and its protein molecular weight varies little from 35.36~47.71 kD.The isoelectric points of these three types of proteins are between 5.25 and8.90 were predicted;Gene structure and conserved domain analysis revealed that motif 1,motif 2,motif 3,motif 4,motif 5,motif 6,motif 9 and motif 10 were shared by most genes,and motif 8 was unique to GA2ox.The 23 gibberellin oxidase genes were clustered on four branches.Transcriptome analysis showed that 6 DEGs in the shoot tips of the tall stem and short stem flax during the rapid growth period,namely 4 GA2ox(LuGA2ox5,LuGA2ox6,LuGA2ox10,LuGA2ox11) and 2 GA20ox(LuGA20ox5,LuGA20ox7).Combined with specific expression analysis of each gene in flax tissue,it was suggested that specific expression of LuGA20ox7 in the shoot tip might be related to the plant height in the vegetative growth period.This study provides a theoretical reference to analyzing the structural characteristics of GAox and the pattern of expression of the growth and development of flax.
