摘 要:通过对肺形侧耳(Pleurotus pulmonarius)杂交后代群体进行农艺性状的综合评价和遗传分析,为肺形侧耳杂交品种选育和应用提供指导。应用灰色关联度分析法对肺形侧耳杂交后代群体及其亲本进行综合评价分析,并利用序列相关扩增多态性(SRAP)标记技术进行杂交菌株及其亲本的遗传分析。结果表明,在杂交后代群体中菌株H155和H232的关联度排名高于亲本,综合农艺性状表现最好,综合评估结果与杂交后代实际栽培表现相一致。18对SRAP引物共扩增出218个条带,其中多态性条带总共为147个,占总条带数的67.43%,有效等位基因数Nei’s基因多态性指数和Shannon信息指数的平均值分别为1.384 1、0.224 9和0.338 9。供试菌株间遗传相似系数(GS)值变化范围为0.63~0.93,在遗传相似系数为0.77时,可将亲本菌株及其杂交后代群体分为Ⅳ个类群,供试菌株在DNA水平上存在一定的遗传差异,SRAP适用于肺形侧耳单孢杂交后代群体的鉴别。研究结果为肺形侧耳杂交后代的筛选及遗传分析提供了一定的理论依据和技术参考。The main purpose of the study is to provide academic references for Pleurotus pulmonarius hybrid breeding and extension by comprehensive evaluation and genetic analysis.Agronomic traits of the Pleurotus pulmonarius hybrids and their parents were comprehensively evaluated by grey relational analysis method,and genetic relationship amongst them was unraveled by sequence-related amplified polymorphic(SRAP) marker technique.The results showed that the correlation degrees of H155 and H232 were ranked higher than the parents in the Pleurotus pulmonarius hybrids and the comprehensive agricultural characteristics were performed best.The comprehensive assessment result was consistent with the actual cultivation performance of the hybrids.The SRAP genotyping suggested that of the 218 fragments produced by 18 SRAP primer combinations 147 were polymorphic,accounting for67.43% of the total number of bands.The effective number of alleles was 1.384 1,the diversity index of Nei's gene was 0.224 9 and the Shannon's information index was 0.338 9.The genetic similarity coefficients of the experimental strains were between 0.62~0.95.With the genetic similarity coefficient of 0.77 as the boundary,15 experimental strains were divided into Ⅳ groups.The selected SRAP primer combinations can be used for the genetic analysis and variety identification of Pleurotus pulmonarius hybrids.The results of the study provide a theoretical basis and technical reference for the screening and genetic analysis of Pleurotus pulmonarius hybrids.
