摘 要:目的观察丙酮酸钠溶液灌胃治疗糖尿病大鼠深Ⅱ°烧伤的效果,并分析可能机制。方法50雄性SD大鼠随机分为对照组、模型组和实验1、2、3组,每组10只;模型组和各实验组采用腹腔注射链脲佐菌素制作糖尿病大鼠模型,造模成功后,于大鼠背部制备一直径4 cm的深Ⅱ°烧伤;实验1、2、3组分别给予2.5%、7.5%、10%的丙酮酸钠溶液灌胃,对照组和模型组给予等量生理盐水灌胃,每天1次。分别于给药1、3、7、14、21 d观察并计算创面愈合率,检测大鼠体质量、血糖,采用ELISA法检测创面组织中的烟酰胺腺嘌呤二核苷酸(NAD+/NADH)、丙二醛(MDA)、超氧化物歧化酶(SOD)、白细胞介素6(IL-6)、肿瘤坏死因子α(TNF-α)。结果模型组给药14、21 d创面愈合率低于对照组,各实验组给药14、21 d创面愈合率高于对照组,且实验3组高于实验1、2组(P均<0.05)。模型组给药各时点血糖均高于对照组,各实验组血糖低于模型组,且实验3组低于实验1、2组(P均<0.05)。模型组给药3~21 d体质量低于对照组、各实验组体质量高于模型组,且实验3组给药7~21 d体质量高于实验1、2组(P均<0.05)。模型组给药后创面组织NAD+/NADH、SOD水平低于对照组,各实验组高于模型组,且实验3组高于实验1、2组(P均<0.05);模型组给药后创面组织MDA、IL-6、TNF-α水平高于对照组,各实验组低于模型组,且实验3组低于实验1、2组(P均<0.05)。结论丙酮酸钠溶液灌胃能够促进糖尿病深Ⅱ°烧伤大鼠创面愈合,降低血糖,维持正常体质量,其机制可能与抑制创面炎症反应及过氧化损伤、改善能量代谢有关。Objective To observe the therapeutic effect of intragastric administration of sodium pyruvate solution on deepⅡ°scald in diabetic rats and to analyze the possible mechanism.Methods Fifty male SD rats were randomly divided into the control group,model group and experimental groups 1,2 and 3,with 10 rats in each group.The diabetic rat models were established by intraperitoneal injection of streptozotocin.After successful modeling,a deep partial thickness scald with diameter of 4 cm was inflicted on the back of each rat in the model group and the experimental groups.Rats in the experimental groups 1,2 and 3 were given 2.5%,7.5%and 10%sodium pyruvate solution by gavage,respectively,once a day,and rats in the control group and the model group were given the same amount of normal saline by gavage.On day 1,3,7,14,and 21 of drug administration,the wound healing rate was observed and calculated,and the body weight and blood glucose of rats were detected.Nicotinamide adenine dinucleotide(NAD+/NADH),malondialdehyde(MDA),superoxide dismutase(SOD),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)in the wound tissues were detected by ELISA.Results The wound healing rates of the model group on day 14 and 21 of administration were lower than those of the control group,and the wound healing rate of each experimental group on day 14 and 21 of administration was higher than that of control group,and the wound healing rate of the experimental group 3 was higher than those of the experimental groups 1 and 2(all P<0.05).The blood glucose of the model group was higher than that of the control group at each time point of administration,and the blood glucose of each experimental group was lower than that of the model group,and that of experimental group 3 was lower than those of experimental groups 1 and 2(all P<0.05).The body weight of the model group was lower than that of the control group from day 3 to 21 of administration,and the body weight of each experimental group was higher than that of the model group,and the body we
