高效离子交换色谱法测定一种^(13)C-尿素呼气试验用药品中^(13)C-尿素的含量

摘　　要：建立采用磺酸基阳离子交换键合硅胶的离子交换色谱法,对^(13)C-尿素呼气试验用药品中^(13)C-尿素的含量进行测定。色谱柱为资生堂CAPCELL PAK SCX UG80S5(4.6 mm×250 mm);流动相为水;流速为0.6 mL/min;柱温为25℃;二极管阵列检测器,检测波长为200 nm,进样量为20μL。结果表明:^(13)C-尿素在0.25~0.60 mg/mL呈良好的线性关系(R=0.9996)。加标回收率为98.1%~100.9%,RSD为1%(n=9)。本方法专属性强、灵敏度高,能够准确地测定^(13)C-尿素的含量,以及控制^(13)C-尿素呼气试验用药品质量。OBJECTIVE To establish a method for determining content of^(13)C-Urea in a drug for^(13)C-UBT by ion used exchange chromatography of sulfonic cation exchange bonded silicone.METHODS SHISEIDO CAPCELL PAK SCX UG80S5(4.6 mm×250 mm)column were adopted,with water as mobile phase at a flow rate of 0.6 mL/min,the column temperature was 25℃,the photo-diode array detector was used with the detection wavelength of 200 nm.And the injection volume was 20μL.RESULTS There was a good linear relationship of^(13)C-Urea in the concentration of 0.25~0.60 mg/mL and the correlation coefficient was 0.9997.The recovery ranged was 98.1%~100.9%with RSD 1%(n=9).The method is specific and sensitive,can accurately determine^(13)C-Urea content and control the quality of drugs for^(13)C-UBT.
