摘 要:人兽共患病是一类可以从动物向人类传播的感染性疾病,约60%的人类感染为动物源性,目前全球已知有250余种人兽共患病,最不发达国家20%的人体疾病和死亡归因于人兽共患病。人兽共患病对人类和动物健康均造成了严重危害,是人类社会经济发展的持续挑战。网络药理学是一门基于系统生物学理论、生物系统网络分析和多靶点药物分子设计特异性信号节点选择的新兴学科,通过整合计算机分析、体内和体外实验及大量数据,发现新的药物靶点、揭示药物作用分子机制。目前,网络药理学已广泛用于临床医学、生命科学、药学、中医药等多个学科领域,对于揭示复杂疾病发病机制、挖掘药物靶标、阐明药物作用机制等均发挥了重要作用。本文主要就网络药理学在人兽共患病研究领域中的应用进展进行综述,从而为人兽共患病防控提供借鉴。Zoonosis is a class of infectious diseases that can be transmitted from non-human animals to humans.Currently,approximately 60%of human infections are zoonotic,and there are 250 known zoonoses across the world,with 20%of human diseases and deaths attributed to zoonoses in the least developed countries.Zoonotic diseases pose great threats to both human and animal health,which are a great challenge in the socioeconomic development in the world.Network pharmacology,an emerging discipline based on systems biological theory,biological system network analysis and multi-target drug molecule design-specific signal nodes selection,combines computational analysis,in vivo and in vitro assays and a large number of data to discover novel drug targets and unravel molecular mechanisms of agents.Network pharmacology has been widely applied in clinical medicine,life sciences,pharmacology and traditional Chinese medicine,which plays important roles in unraveling the pathogenesis of complex diseases,discovering drug targets and elucidating mechanisms of drug actions.This review summarizes the advances in the application of network pharmacology in zoonoses research,so as to provide insights into the management of zoonoses.
