摘  要:目的采用超高效液相-四级杆-飞行时间质谱法(UPLC-Q-TOF-MS/MS)快速鉴定苍术-香附醇提液化学成分和HPLC同时测定苍术-香附醇提液中的6种成分。方法采用UPLC-Q-TOF-MS/MS技术,借助PeakView分析软件,分别在正、负离子模式下定性分析药液中的化学成分。采用HPLC同时定量测定苍术-香附醇提液中的6种成分(木犀草素、白术内酯Ⅰ、白术内酯Ⅱ、白术内酯Ⅲ、α-香附酮、苍术素)。结果共鉴定出54个化合物,包括19个萜类、11个有机酸类、6个萜苷类、6个黄酮类、3个香豆素类、2个聚乙炔类、2个苯丙醇苷类、2个黄酮苷类、1个生物碱类及2个其他类,并对化合物的药材来源进行归属。定量测定的6种成分在各自范围内线性关系良好(r>0.9990),平均加样回收率98.51%~102.38%,RSD 1.879%~4.450%。结论该方法稳定可靠,可用于苍术-香附醇提液的定量及定性检测。OBJECTIVE To establish UPLC-Q-TOF-MS/MS method for rapidly identifing the constituents in alcohol extract of Atractylodes-Cyperus and HPLC method for simultaneously determing six constituents.METHODS Using UPLC-Q-TOF-MS/MS technology,with the help of PeakView software,the chemical components in the liquid medicine were qualitatively analyzed in positive and negative ion modes respectively.Six components(luteolin,atractylideⅠ,atractylideⅡ,atractylideⅢ,α-cyperone,atractylodes)in the alcoholic extract of Atractylodes-Cyperus were quantitatively determined by HPLC method.RESULTS A total of 54 compounds were identified,including 19 terpenoids,11 organic acids,6 terpenoid glycosides,6 flavonoids,3 coumarins,2 polyacetylenes,2 phenylpropanol glycosides,2 flavonoid glycosides,1 alkaloid and 2 other classes.The source of the compounds was identified.Six components showed good linear relationship in their own ranges(r>0.9990),the average recovery were 98.51%-102.38%with the RSDs of 1.88%-4.45%.CONCLUSION The method is stable and reliable,and can be used for quality control for alcohol extract of Atractylodes-Cyperus.
