摘 要:目的建立血瘀证动物模型,构建血瘀证动物模型更加系统、全面的评价体系。方法SPF级雄性SD大鼠随机分为正常组和角叉菜胶(carrageenan,Ca)低、中、高剂量(5、10、20 mg/kg)模型组,模型组分别腹腔注射相应剂量Ca,正常对照组腹腔注射等体积0.9%氯化钠溶液。在造模后的24 h和48 h,对各组大鼠的一般情况、黑尾长度、尾端血流灌注量、舌象、血液流变学、血小板聚集率进行观察和检测,同时留取各组大鼠尾组织进行病理切片分析。结果中剂量组及高剂量组大鼠在造模后24 h和48 h出现活动减少,进食减少,皮毛疏松无光泽,毛发直立,耳郭脉络紫暗等体征。造模后24 h,模型低、中、高剂量组均出现黑尾,高剂量组较低剂量组黑尾长度显著增长(P<0.01);造模后48 h,中剂量组和高剂量组较低剂量组黑尾长度明显增长(P<0.01或P<0.05)。造模后24 h及48 h,模型组各剂量组较正常组尾端血流量显著降低(P<0.01),模型各组之间差异无统计学意义。模型中剂量组及高剂量组在造模后24 h及48 h舌象R、G、B值较正常组显著下降(P<0.01或P<0.05)。造模后24 h及48 h,中剂量组和高剂量组的全血黏度(低、中、高切)显著升高(P<0.01或P<0.05)。造模后24 h及48 h,中剂量组和高剂量组的血小板最大聚集率较正常组显著升高(P<0.01或P<0.05)。尾组织病理显示低剂量组可见少量炎性细胞;中剂量组和高剂量组可见大量炎性细胞浸润,组织间隙水肿,管壁较薄,红细胞溢出。结论中剂量(10 mg/kg)及高剂量(20 mg/kg)角叉菜胶诱导的大鼠血瘀证模型在造模后24 h和48 h呈现更稳定、更典型的血瘀征象;构建了涉及中医血瘀证候宏观表征、现代医学微观指标及组织病理形态学的模型评价体系。Objective To establish an animal model of blood stasis syndrome and to construct a more systematic and comprehensive evaluation system for animal models of blood stasis syndrome.Methods SPF male SD rats were randomly divided into normal group and carrageenan(Ca) low,medium,and high dose(5,10,20 mg/kg) model group;the model group was injected with the corresponding dose of carrageenan in the peritoneum and the normal control group was injected with 0.9% sodium chloride solution.At 24 h and 48 h after injection,the general condition,black tail length,caudal blood perfusion,tongue,blood rheology,and platelet aggregation rate were observed and detected in each group of rats,and the tail tissues of each group of rats were retained for pathological section analysis.Results Rats in the medium and high-dose groups showed signs such as decreased activity,decreased eating,loose and dull fur,erect hair,and purple and dark auricular veins at 24 and 48 hours after molding.24 h after molding,the model low,medium,and high-dose groups all showed off black tails,and the length of the black tail in the high-dose group increased significantly compared with the lowdose group(P<0.01);48 h after molding,compared with the low-dose group,the length of the black tail in the mediumdose and high-dose groups increased significantly(P<0.01 or P<0.05).24 h and 48 h after molding,the tail-end blood flow of each dose group of the model group was significantly reduced compared with the normal group(P<0.01),and no significant difference was found between the model groups.In the medium-dose group and high-dose group of the model,the values of tongue images R,G,and B decreased significantly compared with the normal group 24h and 48h after molding(P<0.01 or P<0.05).The whole blood viscosity(low,medium,and high cut) was significantly increased in the mediumand high-dose groups at 24 and 48 hours after molding(P<0.01 or P<0.05).At 24 and 48 h after molding,the maximum aggregation rate of platelets in the medium-dose and high-dose groups was significan
