摘　　要：建立了高效液相色谱-蒸发光散射测定虫草头孢菌粉中D-甘露醇的分析方法。样品经过超声波辅助提取后,以Shodex Asahipak NH2P-504E(4.6 mm×250 mm,5μm)为分离柱,乙腈-水为流动相,梯度洗脱,流速1.0 mL/min。蒸发光散射检测器参数:漂移管温度35℃,氮气流速1.60 SLM。在0.100~2.00 g/L范围内D-甘露醇质量浓度的自然对数与峰面积的自然对数呈良好线性。相对标准偏差为5.8%,加标回收率为93.5%~97.5%。该方法基质干扰较小,简单快速,结果准确可靠和灵敏度高,适用于虫草头孢菌粉中D-甘露醇含量的测定。To establish a high performance liquid chromatography with evaporative light scattering detection(ELSD)method for the determination of D-mannitol in cordyceps cephalosporium mycelia,the sample was treated by ultrasonic assisted extraction and then was performed on a Shodex Asahipak NH2P-504E column(4.6 mm×250 mm,5μm).The mixture of acetonitrile and water was chosen as the mobile phase under a gradient elution at a flow rate of 1.0 mL/min.The ELSD parameters were as follows:drift tube temperature was 35℃ and nebulizing gas flow rate was 1.60 SLM.The natural logarithm of D-mannitol mass concentration had a good linearity with the natural logarithm of the peak area in the range of 0.100~2.00 g/L.The relative standard deviation was 5.8%and the recoveries were between 93.5%~97.5%.This method was found to be less interference of matrix,simple,rapid,accurate and sensitive,and can be suitablefor the determination of D-mannitol in cordyceps cephalosporium mycelia.
