摘 要:目的以正常SD大鼠为研究对象,探究枣仁安神颗粒对其睡眠—觉醒规律的影响。方法将8只雄性SD大鼠进行有创脑电(electroencephalography,EEG)和肌电(electromyography,EMG)的埋置手术后,单笼饲养7天进行术后恢复;术后第8天开始上机进行无线遥感监测睡眠—觉醒,光照和黑暗为12小时自然交替,以保证其正常昼夜节律,连续监测3天;随后给予枣仁安神颗粒,按874.05 mg/(kg·d)、水33 mL/d,药水混合自由取用,连续给药8天;从给药的第6天开始,上机监测睡眠—觉醒连续3天;结束后导出EEG、EMG数据,通过EDFbrowser软件对记录的数据进行睡眠—觉醒分析,并导出分析结果;使用IBM SPSS Statistics 25统计分析。结果与正常睡眠相比,给予枣仁安神颗粒后的大鼠睡眠期总时长明显延长(P<0.05),清醒期时长明显缩短(P<0.05);其中,非快速眼动睡眠期时长显著增加(P<0.05),快速眼动睡眠期时长显著增加(P<0.05)。结论在觉醒期,EEG呈高频率,低振幅,EMG呈高频率的肌肉收缩;在非快速眼动睡眠期时,EEG呈高低振幅交替的不规则图像,呈慢波形式;在快速眼动睡眠期时,EEG呈低幅快波形式。枣仁安神颗粒对正常大鼠睡眠—觉醒规律有干预作用,大鼠清醒时长缩短,睡眠时间延长,对非快速眼动睡眠期和快速眼动睡眠期均具有延长作用。Objective To investigate the effect of Zaoren Anshen granule on sleep-wake pattern in normal SD rats.Method Eight male SD rats were subjected to invasive electroencephalography(EEG)and electromyography(EMG)implantation surgery,and were housed in a single cage for 7 days for postoperative recovery;on the 8th day after surgery,wireless remote sensing was performed to monitor sleep-wake on the machine,and light and darkness were naturally alternated for 12 hours to ensure their normal circadian rhythm for 3 days;then Zaoren Anshen granules were given,according to 874.05 mg/(kg·d),water 33 mL/d,and the liquid mixture was freely available for 8 days;from the 6th day of administration,sleep-wake was monitored on the machine for 3 days;EEG and EMG data were exported after the end,and sleep-wake analysis was performed on the recorded data by EDFbrowser software,and the analysis results were exported;IBM SPSS Statistics 25 statistical analysis was used.Result Compared with normal sleep,the total sleep duration of rats given Zaoren Anshen granule was significantly increased(P<0.05),and the awake duration was significantly decreased(P<0.05);among them,the non-rapid eye movement(NREM)duration was significantly increased(P<0.05),and the rapid eye movement(REM)duration was significantly increased(P<0.05).Conclusion During the awakening period,EEG exhibits high frequency and low amplitude,while EMG exhibits high frequency muscle contraction;during non rapid eye movement sleep,EEG showed irregular images with alternating high and low amplitudes,in the form of slow waves;during rapid eye movement sleep,EEG showed low radiation and fast wave.Zaoren Anshen granule can intervene the sleep-wake regularity of normal rats,decrease the awake time and increase the sleep of rats,and increase the non-rapid eye movement sleep period and rapid eye movement sleep period.
