HFPO-DA染毒对孕鼠甲状腺内分泌功能的影响

摘  要:目的探讨孕期染毒六氟环氧丙烷二聚体羧酸(HFPO-DA)对孕鼠甲状腺内分泌功能的影响。方法将妊娠0.5 d的SD孕鼠随机分为对照组(去离子水)和HFPO-DA低剂量(1 mg/kg)、中剂量(10 mg/kg)、高剂量组(100 mg/kg),每组8只。妊娠0.5~19.5 d,每天按体质量进行染毒。在妊娠19.5 d用100 g/L水合氯醛麻醉后断头处死所有孕鼠,检测各组胎鼠的生长发育指标,苏木精-伊红染色观察孕鼠甲状腺组织形态,酶联免疫吸附方法检测孕鼠血清促甲状腺激素(TSH)、总四碘甲状腺原氨酸(TT_(4))和总三碘甲状腺原氨酸(TT_(3))的浓度,高效液相色谱-串联质谱法检测孕鼠血清HFPO-DA浓度。结果与对照组比较,高剂量HFPO-DA染毒可显著降低胎鼠的出生体质量、身长和尾长,显著升高孕鼠TSH和TT_(3)的水平,差异有显著性(t=5.07~38.64,P<0.05),并且导致孕鼠甲状腺组织出现增生小滤泡。结论高剂量HFPO-DA染毒可以损伤孕鼠甲状腺结构,干扰孕鼠体内甲状腺激素的水平,进而导致胎鼠宫内生长发育落后。Objective To investigate the effects of exposure to hexafluoropropylene oxide dimer acid(HFPO-DA)on thyroid endocrine function in pregnant rats.Methods Sprague-Dawley(SD)rats(pregnant for 0.5 d)were randomly divided into control group(deionized water)and low-dose(1 mg/kg),medium-dose(10 mg/kg),and high-dose(100 mg/kg)HFPO-DA groups,with eight rats in each group.The rats were exposed to HFPO-DA daily based on body weight during the gestational period of 0.5 to 19.5 d.On day 19.5,all the pregnant rats were decapitated after anesthesia with chloral hydrate at 100 g/L to measure the growth and development data of the fetal rats.Thyroid tissue morphology was observed with hematoxylin-eosin staining.The se-rum levels of thyroid stimulating hormone(TSH),total thyroxine(TT_(4)),and total triiodothyronine(TT_(3))in the pregnant rats were measured by enzyme-linked immunosorbent assay.Serum HFPO-DA levels were determined by high performance liquid chromatography tandem mass spectrometry.Results Compared with the control group,exposure to high-dose HFPO-DA significantly decreased fetal rats’body weight,body length,and tail length at birth,and significantly increased the levels of TSH and TT_(3)in pregnant rats(t=5.07-38.64,P<0.05),with follicular hyperplasia present in the thyroid tissue.Conclusion High-dose HFPO-DA exposure can damage the thyroid structure and disrupt the thyroid hormones of pregnant rats,leading to intraute-rine growth and development retardation of offspring.
