摘  要:目的探究丹参-葛根提取物(DG)对动脉粥样硬化(AS)小鼠的保护作用。方法选择6周龄的ApoE-/-雄性小鼠24只,随机分为空白组、模型组、DG低剂量组(200 mg/kg)和DG高剂量组(400 mg/kg),每组6只。空白组用正常饲料喂养,模型组和DG治疗组用高糖高脂饲料喂养;空白组和模型组每天灌胃生理盐水,DG治疗组每天灌胃DG提取液,实验持续16周。实验结束后应用生化试剂盒检测小鼠血清中脂质成分三酰甘油(TG)、总胆固醇(TC)和低密度脂蛋白(LDL-C)水平,氧化应激因子丙二醛(MDA)、超氧化物歧化酶(SOD)、谷胱甘肽(GSH)的含量;采用油红O染色和Masson染色观察小鼠主动脉病理组织学变化;采用ELISA方法检测血清中炎症因子肿瘤坏死因子α(TNF-α)、白细胞介素12(IL-12)、可溶性细胞间黏附分子1(sICAM-1)、可溶性血管细胞黏附分子1(sVCAM-1)水平。结果生化检测结果显示,与空白组比较,模型组小鼠血清中的TG、TC和LDL-C水平显著升高,给予药物DG干预后可以浓度依赖性改善脂质紊乱(F=180.0~564.6,P<0.001);模型组小鼠血清中的氧化应激因子MDA含量显著升高,抗氧化因子SOD、GSH含量显著降低,给予DG干预后MDA血清含量有效降低,抗氧化因子SOD、GSH含量升高(F=133.0~194.7,P<0.001)。ELISA检测结果显示,与正常小鼠相比,模型组小鼠血清中炎症因子TNF-α和IL-12以及sICAM-1、sVCAM-1水平显著增加,DG剂量依赖性抑制炎症因子水平(F=247.0~371.0,P<0.001)。油红O和Masson染色结果显示,与模型组比较,DG低剂量和高剂量组小鼠动脉斑块面积减少,斑块处胶原纤维含量增加。结论DG能降低AS小鼠脂质过氧化水平,抑制炎症反应,减少斑块的面积,提高易损斑块的稳定性,抑制AS的发生和发展。Objective To investigate the protective effects of Salvia miltiorrhiza Bunge-Radix Puerariae extract(DG)in mice with atherosclerosis(AS).Methods Twenty-four ApoE-/-mice(6 weeks old,male)were randomly divided into blank group,model group,low-dose DG group(200 mg/kg),and high-dose DG group(400 mg/kg),with six mice in each group.The mice in the blank group were fed a normal diet,while those in the model group and DG treatment groups were fed a high-sugar and high-fat diet.The blank and model groups were intragastrically given normal saline,while the DG groups received intragastric injection of DG,for a continuous period of 16 weeks.The serum levels of lipids(triglyceride(TG),total cholesterol(TC),and low-density lipoprotein cholesterol(LDL-C))and oxidative stress factors(malondialdehyde(MDA),superoxide dismutase(SOD),and glutathione(GSH))were measured using biochemical kits.Serum inflammatory factors(tumor necrosis factor-α(TNF-α),interleukin-12(IL-12),soluble intercellular adhesion molecule-1(sICAM-1),and soluble vascular cell adhesion molecule-1(sVCAM-1))were measured by enzyme-linked immunosorbent assay(ELISA).The pathological changes of the aorta were observed with oil red O staining and Masson staining.Results Compared with those in the blank group,serum TG,TC,and LDL-C levels in the model group were significantly increased;DG treatment significantly ameliorated dyslipidemia in a dose-dependent manner(F=180.0-564.6,P<0.001).The model group showed a significantly increased serum MDA level and significantly decreased serum SOD and GSH levels;DG treatment significantly reduced serum MDA levels and increased SOD and GSH levels(F=133.0-194.7,P<0.001).The inflammatory factors TNF-α,IL-12,sICAM-1,and sVCAM-1 were significantly increased in the model group than in the normal mice;and these indicators were significantly inhibited by DG treatment in a dose-dependent manner(F=247.0-371.0,P<0.001).Oil red O staining and Masson staining results showed that compared with the model group,the low-dose and high-dose DG group
