摘  要:目的探索三氧化二铁纳米颗粒(Fe_(2)O_(3)NPs)经口暴露后在大鼠小肠中的吸收转运特点及机制。方法26只雄性SD大鼠按体重随机分为吸收曲线组和重复灌胃组[对照组(超纯水);实验组200 mg/(kg·bw)]。前者采眼眶血测定铁离子含量并绘制吸收曲线计算药动学参数。重复灌胃组连续灌胃28天后处死大鼠采集胃液、肠液后进行体外模拟消化实验、检测转运相关基因表达、铁离子含量和纳米颗粒分布情况。结果Fe_(2)O_(3)NPs在人工模拟胃肠液及大鼠胃肠液中释放的铁离子随消化时间延长而增加且符合二室曲线代谢模型。与对照组相比,实验组大鼠各肠段铁离子转运相关基因和肠上皮胞吞相关基因表达升高(P<0.05);各肠段铁离子含量升高(P<0.05),且各肠段铁离子含量和纳米颗粒的沉积数量均表现为十二指肠>空肠>回肠。结论本研究中,单次灌胃Fe_(2)O_(3)NPs后大鼠的口服吸收率低,消除较快。Fe_(2)O_(3)NPs在大鼠小肠的吸收方式包括以铁离子和以纳米颗粒形式肠上皮细胞胞吞,且发生在整个小肠。Objective To investigate the absorption and transport characteristics and mechanisms of ferric oxide nanoparticles(Fe_(2)O_(3)NPs)in the small intestine of rats following oral exposure.Methods Twenty-six male Sprague-Dawley(SD)rats were randomly divided into two groups:the absorption curve group and the repeated gavage group.[control group(ultrapure water);experimental group 200 mg/(kg·bw)]according to body weight.In the former group,orbital blood was collected to determine the iron ion content,and absorption curves were plotted to calculate pharmacokinetic parameters.After repeated gavage for 28 days,the rats were executed to collect gastric and intestinal fluids.Subsequently,in vitro digestion experiments were performed to assess the expression of transport-related genes,iron ion content and nanoparticle distribution.Results The release of Fe_(2)O_(3)NPs in artificial simulated gastrointestinal fluid and rat gastrointestinal fluid increased with prolonged digestion time,exhibiting conformity with a two-compartment curve metabolic model.Compared with the control group,the expression of iron ion transport-related genes and intestinal epithelial cytophagy-related genes was increased in all intestinal segments of rats(P<0.05);the iron ion content of each intestinal segment was increased,and the iron ion content and the number of nanoparticles deposited in each intestinal segment showed that duodenum>jejunum>ileum(P<0.05).Conclusion This study reveals a low oral absorption rate of Fe_(2)O_(3)NPs in rats after a single gavage,accompanied by rapid elimination.The absorption of Fe_(2)O_(3)NPs in the small intestine of rats occurs in the form of iron ions and nanoparticles,involving intestinal epithelial cell cytokinesis,and is distributed throughout the entire small intestine.
