摘　　要：以聚乙烯醇(PVA)和壳聚糖(CS)为原料,采用钴-60γ射线辐射制备了PVA/CS复合水凝胶。考察了该复合水凝胶溶胀行为和透明性,并通过扫描电子显微镜(SEM)对水凝胶的微观形貌进行了表征;开展了PVA/CS复合水凝胶的刺激与皮肤致敏试验、全身毒性试验和溶血试验等生物相容性评价。结果表明:将CS引入PVA水凝胶后,PVA/CS复合水凝胶不仅具有良好的透明性,而且达到溶胀平衡时的溶胀度比纯PVA水凝胶提高了7.8倍,达到了248%;与纯PVA水凝胶相比,PVA/CS复合水凝胶网络中的孔洞大小和深度均有所增加。PVA/CS复合水凝胶不仅对兔和豚鼠的皮肤刺激性极小且无致敏反应,而且在浸提液中并无有毒物质析出,对小鼠无急性毒性作用。PVA/CS复合水凝胶溶血率仅为2.6%,具有良好的血液相容性。Polyvinyl alcohol(PVA)and chitosan(CS)were used as raw materials to prepare PVA/CS composite hydrogel by Co-60γ-ray radiation.The swelling behavior and transparency of the composite hydrogels were investigated.The microscopic morphology was characterized by scanning electron microscopy(SEM).The biocompatibility of the hydrogel was evaluated through irritation and skin sensitizing test,systemic toxicity test and hemolysis test.The results showed that after the introduction of CS into the PVA hydrogel,the PVA/CS hydrogel had good transparency,and the equilibrium swelling degree reached 248%,which was 7.8 times higher than that of pure PVA hydrogel.The pore size and depth in the PVA/CS composite hydrogel network were increased compared with pure PVA hydrogel.The PVA/CS composite hydrogel exhibited minimal skin irritation and no sensitization reaction to rabbits and guinea pigs.Additionally,no toxic substances were released in the extract,and the hydrogel had no acute toxicity to mice.The hemolysis rate of PVA/CS composite hydrogel was only 2.6%,indicating good blood compatibility.
