摘  要:试验旨在优化白头翁口服液的提取和澄清工艺。采用L9(34)正交试验对白头翁口服液的水提与澄清工艺进行优化。试验固定提取次数3次,以液料比、浸泡时间、提取时间为考察因素,以盐酸小檗碱含量为评价指标,对水提工艺进行探索研究;以药液浓度、澄清剂用量、絮凝温度为考察因素,以吸光度、盐酸小檗碱保留率为综合评价指标,对澄清工艺进行探索研究,同时对絮凝澄清工艺与醇沉澄清工艺进行比较。结果显示,白头翁口服液的最佳水提工艺为液料比10 mL/g、浸泡时间30 min、提取次数3次、每次提取时间70 min。最佳澄清工艺为药液浓度1∶7 g/mL,ZTC1+1天然澄清剂用量6%A、3%B,絮凝温度60℃。絮凝澄清工艺除杂后盐酸小檗碱平均保留率为70.89%,而醇沉澄清工艺的盐酸小檗碱平均保留率仅56.72%,且不同批次絮凝澄清制备口服液测定的含量保留率相对标准偏差(RSD)值<1.00%(n=3)。研究表明,试验优化的提取澄清工艺稳定可靠,可用于工业化生产。The study aimed to optimize the extraction and clarification process of Baitouweng oral liquid.The L9(34)orthogonal test was used to optimize the water extraction and clarification process of Baitouweng oral liquid.The extraction times were fixed for three times,the liquid to solid ratio,soaking time and extraction time were considered as factors,and the content of berberine hydrochloride was taken as evaluation index to explore the water extraction process.With the concentration of liquid,dosage of clarifying agent and flocculation temperature as factors,the absorbance and retention rate of berberine hydrochloride as comprehensive evaluation indexes,the clarifying process was explored and studied.At the same time,the flocculation clarifying process was compared with alcohol precipitation clarifying process.The results showed that the optimal water extraction process of Baitouweng oral liquid was as follows:liquid to solid ratio 10 mL/g,soaking time 30 min,extraction for three times,each extraction time 70 min.The optimal clarifying process was as follows:liquid concentration 1∶7 g/mL,dosage of ZTC1+1 natural clarifying agent 6%A and 3%B,flocculation temperature 60℃.The average retention rate of the berberine hydrochloride was 70.89%after flocculation and clarification process,while it was only 56.72%for berberine hydrochloride using the alcohol precipitation method.The relative standard deviation(RSDS)of content retention rate of the oral liquid prepared by flocculation and clarification from different batches was lower than 1.00%(n=3).The study indicates that the extraction and clarification process optimized in this experiment is stable and reliable.It can be used in industrial production.
