刺五加注射液对模拟失重大鼠心血管功能调节作用研究

摘  要:目的空间失重明显影响航天员心血管系统的正常生理功能,传统中药以整体综合调节发挥潜力,研究传统中药刺五加注射液对模拟失重大鼠心血管功能的保护作用,以改善空间失重环境下的心血管功能。方法采用头低位尾吊大鼠模拟失重,动态超声、心肌酶谱、血清钙离子浓度及心肌相关mRNA和蛋白质表达指标研究失重对心血管功能的影响及刺五加注射液的干预机理。结果与对照相比,模拟失重使大鼠心率显著升高约5%,心室壁厚度、内径、每搏输出量、心排量以及血清Ca^(2+)均有不同程度的减小,左室射血分数和收缩率升高。心肌酶谱中CK和CKMB分别升高33%和42%。对信号通路Ca^(2+)/CaN/NFAT3下游基因Myh6、Myh7、ANF上调,BNP、CaN、α-actin下调,对蛋白质CaN、NFAT3表达量上调,P-NFAT3下调。结论刺五加注射液通过增加失重大鼠心室厚度、每搏输出量和心排量和细胞钙排量,降低心率和收缩率,调节Ca^(2+)/CaN/NFAT3信号通路基因及蛋白表达,发挥心血管保护作用,高剂量药效较好。Objective Space weightlessness obviously affects the normal physiological function of astronauts’cardiovascular system.Traditional Chinese medicine plays its potential by overall comprehensive regulation.To study the protective effect of traditional Chinese medicine acanthopanax senticosus injection(ASI)on cardiovascular function in rats with simulated weight loss,so as to improve the cardiovascular function in the environment of space weightlessness.Methods The effects of weightlessness on cardiovascular function and the intervention mechanism of ASI were studied by ultrasound,myocardial enzyme,serum calcium concentration and mRNA and protein expression of myocardium in rats with low head and tail suspension.Results Compared with the control group,simulated weightlessness significantly increased heart rate by about 5%,decreased ventricular wall thickness,inner diameter,stroke output,cardiac displacement and serum Ca^(2+),and increased left ventricular ejection fraction and systolic rate.CK and CKMB increased by 33%and 42%,respectively.The signaling pathway Ca^(2+)/CaN/NFAT3 downstream genes Myh6,Myh7,ANF were upregulated,while BNP,CaN,α-actin were downregulated,and up-regulation of the proteins CaN and NFAT3 expression,and down-regulation of P-NFAT3.Conclusion ASI can reduce heart rate and systole rate,and regulate Ca^(2+)/CaN/NFAT3 signaling pathway gene and protein expression by increasing ventricular thickness,stroke volume,cardiac displacement and cellular calcium displacement in rats with severe loss,and play a protective role in cardiovascular system.High dose group is more effective.
