摘  要:目的:系统分析冠心丹参滴丸主要成分及其给药后大鼠体内的主要入血成分。方法:采用高效液相色谱-质谱联用(high performance liquid chromatography-mass spectrometry,LC-MS)方法,分别在正离子模式和负离子模式下对冠心丹参滴丸提取物进行一级和二级质谱分析,通过与对照品及文献比对,确定冠心丹参滴丸的主要成分;在此基础上,采用建立的LC-MS方法分析冠心丹参滴丸给药后的大鼠含药血清,通过与空白大鼠血清比较,确定冠心丹参滴丸的入血成分,并进行结构鉴定。结果:从冠心丹参滴丸提取物中检测到28个主要组分,其中人参皂苷类成分13个、醌类6个(丹参)、酚酸类4个(丹参)和异黄酮类5个(降香)。含药血清检测发现,人参皂苷Rb1和Rd、隐丹参酮和丹参酮ⅡA为冠心丹参滴丸给药后在大鼠体内的主要入血成分。结论:人参皂苷类和丹参中醌类成分是冠心丹参滴丸的主要成分和入血的主要成分,该研究为冠心丹参滴丸后期的药效物质基础和质量控制研究奠定基础。OBJECTIVE To investigate the main chemical components and the in vivo absorbed components of Guanxin Danshen Dripping Pills.METHODS A high performance liquid chromatography coupled with tandem mass spectrometry(LC-MS)was applied to analyze and characterize the extract of Guanxin Danshen Dripping Pills.The MS and MS2 data of the main components were analyzed comprehensively.Being compared with the reference standards and literatures,these constituents were identified or tentatively characterized.Then,the medicated serums of Guanxin Danshen Dripping Pills in rats were analyzed by the established LC-MS approach.By compared with the blank serum and the extract of Guanxin Danshen Dripping Pills,the absorbed circulating components of Guanxin Danshen Dripping Pills were screened and identified.RESULTS A total of 28 compounds were tentatively identified from Guanxin Danshen Dripping Pills,including 13 ginsenosides,6 quinones and 4 phenolic acids from Salvia miltiorrhiza,and 5 isoflavones from Dalbergia odorifera.Among these components,ginsenoside Rb1,ginsenoside Rd,cryptotanshinone,and tanshinoneⅡA were found to be the main absorbed circulating components of Guanxin Danshen Dripping Pills in rats.CONCLUSION Ginsenosidesand quinonesconstitute the main components of Guanxin Danshen Dripping Pills and were its major in vivo absorbed components.Ginsenosides and quinones might be the potential bioactive compounds for the pharmacodynamics effects of Guanxin Danshen Dripping Pills.This study is helpful for the quality control of Guanxin Danshen Dripping Pills and is essential to uncover its effective substances.
