摘 要:目的探讨藏药二十五味鬼臼丸对雌性去卵巢骨质疏松大鼠骨代谢平衡的影响。方法将48只3月龄SD雌性大鼠随机分为OVX、Sham、E_(2)、GJW组。用ELISA法检测血清OC、TRAP5b、Ca、E_(2)、LH含量,以TRAP法将股骨远端干骺端染色,通过三点弯曲实验获得骨生物力学相关数据,用扫描电镜观察股骨远端松质骨超微结构。结果与Sham组比较,OVX组大鼠TRAP5b、LH升高(P<0.05,P<0.05),OC、Ca、E_(2)水平降低(P<0.05,P<0.05,P<0.05);破骨细胞数明显上升(P<0.05);骨超微结构破坏明显,骨最大载荷、最大位移明显下降(P<0.05,P<0.05)。与OVX组比较,GJW组和E_(2)组TRAP5b、LH水平下降(P<0.05,P<0.05),OC、Ca、E_(2)水平升高(P<0.05,P<0.05,P<0.05);破骨细胞数明显减少(P<0.05,P<0.05);骨最大载荷、最大位移明显上升(P<0.05,P<0.05);骨胶原纤维走向趋于整齐。结论藏药二十五味鬼臼丸具有雌激素样作用,对雌性去卵巢骨质疏松大鼠骨代谢平衡产生影响:可有效改善雌性去卵巢骨质疏松大鼠骨质疏松症的骨代谢和血清指标及骨微结构,抑制破骨细胞形成,显著提高PMOP大鼠股骨最大载荷及最大位移。Objective This paper aimed to investigate the effects of Tibetan Medicine Ershiwuwei Guijiuwan on bone metabolism balance in Female ovariectomized rats with osteoporosis.Methods Forty-eight 3-month-old SD female rats were randomly divided into OVX,Sham,E_(2) and GJW group.The enzyme-linked immunosorbent assay(ELISA)was performed to detect the serum levels of OC,TRAP5b,Ca,E_2 and LH.TRAP staining was performed to stain the metaphysis of distal femur.Three-point bending experiment was performed to obtain the biomechanical data of bone.Scanning electro-microscopy was performed to observe the ultrastructure of cancellous bone of distal femur.Results Compared with the Sham group,the level of TRAP5b and LH were increased in OVX group(P<0.05,P<0.05),while OC,Ca and E_2 levels were decreased(P<0.05,P<0.05,P<0.05).The number of osteoclasts increased significantly(P<0.05).The ultrastructure of bone was damaged obviously,and the maximum load and displacement of bone were decreased significantly(P<0.05,P<0.05).Compared with the OVX group,the level of TRAP5b and LH in GJW and E_(2) groups were decreased(P<0.05,P<0.05),while OC,Ca and E_2 levels were increased(P<0.05,P<0.05,P<0.05).The number of osteoclasts decreased significantly(P<0.05,P<0.05).The maximum load and displacement of bone increased significantly(P<0.05,P<0.05).The collagen fibrils tend to be neat.Conclusion Tibetan Medicine Ershiwuwei Guijiuwan has estrogenic effects,affecting bone metabolic balance in female ovariectomized osteoporotic rats:it can effectively improve bone metabolism,serum indexes and bone microstructure of osteoporosis in female ovariectomized osteoporotic rats,inhibit osteoclast formation,and significantly increase the maximum load and displacement of femur in PMOP rats.
