摘  要:目的分析青海玛多地区(海拔4300 m)藏族肥胖儿童与正常体质量儿童粪便代谢组分是否不同;差异代谢物组成及相关代谢途径。方法用液相色谱-质谱联用(LC-MS)技术的非靶向代谢组学方法进行代谢物鉴定,所得数据与数据库进行比对。使用SIMCA14.1软件进行主成分分析(PCA)、正交偏最小二乘法-判别分析(OPLS-DA),通过OPLS-DA模型筛选出变量重要性投影(VIP)值>1的差异代谢物,用MetaboAnalyst 5.0软件进行单因素t检验筛选出两组间的差异代谢物,并计算代谢物在两组间的差异倍数(fold change,FC)。最终用VIP、FC、P三个参数的交集确定差异代谢物;通过KEGG数据库对差异代谢物进行通路富集分析。结果(1)通过PCA、OPLS-DA分析法,在整体水平上对代谢物的组间差异进行可视化分析,各组样本代谢物在组内的分布是均匀的,在组间的分布存在差异;(2)在正离子模式下筛选出差异代谢物20个(5个上调,15个下调),在负离子模式下筛选出差异代谢物14个(11个上调,3个下调);(3)叶酸依赖性碳池代谢,柠檬酸循环代谢,乙醛酸和二羧酸代谢,丙氨酸、天冬氨酸和谷氨酸代谢通路显著富集(P<0.05),主要富集的差异代谢物是柠檬酸、α-酮戊二酸。结论(1)青海玛多地区藏族肥胖儿童粪便代谢组分存在差异;(2)大多数差异代谢物以脂质分子为主;(3)可能通过影响柠檬酸循环代谢,乙醛酸、二羧酸代谢,丙氨酸、天冬氨酸、谷氨酸代谢途径影响机体的能量代谢。Objective To analyze the fecal metabolomics of Tibetan obese children and normal body weights children in Maduo area(altitude 4300 m)of Qinghai and to investigate the composition of differential metabolites and metabolic pathways.Methods The metabolites were identified by non-targeted metabolomics using liquid chromatography-mass spectrometry(LC-MS),and the data were compared with databases.Principal component analysis(PCA)and orthogonal partial least squares-discriminant analysis(OPLS-DA)were performed using SIMCA14.1 Differential metabolites with VIP value>1 were screened by OPLS-DA model,Single factor t test was performed by MetaboAnalyst 5.0 to screen out the differential metabolites between the two groups,and the fold change(FC)between the two groups was calculated.VIP value,FC value and P value were used to screen differential metabolites.Pathway enrichment analysis of differential metabolites was performed using KEGG database.Results(1)Through PCA and OPLS-DA analysis methods to visually analyze the differences between groups of metabolites,it can be seen that the distribution of samples in each group is uniform,while the distribution between groups is different;(2)20 differential metabolites were screened in the positive ion mode(5 were up-regulated and 15 were down-regulated).In the negative ion mode,14 differential metabolites were screened(11 were up-regulated and 3 were down-regulated);(3)Folic acid-dependent carbon pool metabolism,citric acid cycle,glyoxylic acid and dicarboxylic acid metabolism,alanine,aspartic acid and glutamic acid metabolism pathway were significantly enriched(P<0.05).Conclusions(1)There are differences in the fecal metabolites of Tibetan obese children in Maduo area of Qinghai;(2)Most differential metabolites are mainly lipid molecules;(3)Energy metabolism may be affected by the citric acid cycle,glyoxylic acid and dicarboxylic acid metabolism,alanine,aspartic acid and glutamic acid metabolism.
