摘 要:F亚属的人类腺病毒(Human adenovirus,HAdV)与急性胃肠炎相关,是儿童急性胃肠炎住院的常见病因。本研究收集了712份<5岁儿童急性胃肠炎腹泻监测粪便标本,检出HAdV-F阳性52例,阳性检出率为7.30%。对Ct值<30的样本进行文库构建及测序,经Blast比对,得到的21条全基因组序列均为HAdV-41。与HAdV-41原型株相比较,Long fiber和Short fiber的核苷酸一致性低于全基因组核苷酸一致性。对HAdV-41全基因组的遗传进化分析显示,其分子进化速率为7.882×10^(-5)替换/位点/年(95%HPD,3.885×10^(-5)~1.234×10^(-4))。本课题研究对南通地区流行的HAdV-41进行了流行病学特征及基因进化特征分析,为南通地区HAdV-41所致疾病防控及病原溯源奠定了基础。Human adenovirus (HAdV) of subgenus F is associated with acute gastroenteritis and is a common cause of hospitalization for acute gastroenteritis in children.In this study,712 stool samples were collected from children<5 years old for acute gastroenteritis diarrhea monitoring,and 52 cases were positive for HAdV-F,with a positive rate of 7.30%.Libraries were constructed and sequenced for samples with a Ct value<30,and21 whole-genome sequences obtained were all HAdV-41 by Blast comparison.Compared with the HAdV-41prototype strain,the nucleotide identity of long fiber and short fiber were lower than that of the whole genome.The genetic evolution analysis of the whole genome of HAdV-41 showed that its molecular evolution rate was7.882×10~(-5) substitution/site/year (95%HPD,3.885×10~(-5)~1.234×10~(-4)).In this study,the epidemiological characteristics and gene evolution characteristics of HAdV-41 prevalent in Nantong area were analyzed,which laid a foundation for prevention and control of diseases caused by HAdV-41 in Nantong area,China and the traceability of pathogens.
