摘  要:钙依赖蛋白激酶(CPK)可作为Ca^(2+)的感受器和效应器,在结合Ca^(2+)后通过解除自抑制而激活自身激酶活性,并通过磷酸化靶蛋白实现信号转导。为了筛选鉴定胡椒CPK基因家族,探讨其基因进化及表达模式,本研究依据已公布胡椒基因组信息,利用生物信息学方法对胡椒CPK基因家族进行鉴定,共鉴定获得30个CPK基因,命名为PnCPK1~PnCPK30,并对该家族基因进行染色体定位、系统进化、基因结构、保守基序、基因复制事件和表达分析。结果显示,大多数PnCPK基因含有6~8个内含子,30个PnCPK基因不均等地分布在18条染色体上,共有21对复制基因对,串联重复仅2对,Ka/Ks结果说明,胡椒CPK基因家族在进化选择中主要受纯化选择影响;为进一步分析胡椒和其他植物的同源进化关系,构建了胡椒与拟南芥、水稻的CPK进化树,胡椒CPK家族可分为4个亚族,GroupⅡ和GroupⅢ较保守;基于转录组数据结果显示,PnCPK家族各成员表达差异较明显,多数PnCPK成员在各组织中表达量差异不大,荧光定量PCR结果推测PnCPK4、PnCPK10和PnCPK16可能参与了低温和Ca^(2+)介导的低温调控。本研究为深入解析PnCPK基因功能及低温胁迫应答反应中的功能提供参考依据。Calcium-dependent protein kinase(CPK) function as Ca^(2+)sensors and effectors to translate Ca^(2+) signals by phosphorylated target protein after releasing its own kinase activity inhibition.In this study,to identify CPK gene family and analyze gene evolutionary and expression patterns,based on the published Piper nigrum genome data,the CPK gene family members of P.nigrum were identified by bioinformatics methods,and a total of 30CPKs were identified,named PnCPK1~PnCPK30,whose chromosomal mapping,phylogeny,gene structure,conserved motifs,gene replication events and expression were analyzed.The results showed that most PnCPK genes contained 6~8 introns.All the PnCPKs were unequally distributed on 18 chromosomes,of which duplicated gene pairs has 21,and tandem repeats only two.The results of Ka/Ks indicated that the PnCPK gene family was mainly affected by purification selection in evolutionary constraints.In order to further analyze the homologous evolutionary relationship between P.nigrum and other plants(Arabidopsis and rice),the evolutionary tree was built.PnCPKs were phylogenetically divided into four subgroups,with the subgroup Ⅱ and Ⅲ were the most conservative.Based on transcriptome data showed that the expression of PnCPK family members was significantly different,and most of them had no significance expression in various tissues.Furthermore,the results of RT-PCR suggested that PnCPK4,PnCPK10 and PnCPK16 might be involved in the regulation of low temperature and Ca^(2+)-mediated low temperature response.This study provides a reference for further analysis of PnCPK gene function and its function in response to cold stress.
