摘  要:为了研究元宝枫种质资源遗传的多样性,本研究对所采集的元宝枫无性系群体DNA进行荧光PCR扩增,利用毛细管电泳法检测PCR产物,分析元宝枫的集中分布区域之间及同源无性系间的遗传差异和遗传多样性。用11对荧光标记SSR引物对250个元宝枫无性系进行分析,数据显示共102个等位基因,每个位点的等位基因数值4~16个不等,平均等位基因数为9.27;观测杂合度(Ho)为0.42~0.94,平均观测杂合度为0.66,期望杂合度(He)为0.45~0.88,平均期望杂合度为0.73。Shannon’s多样性指数(I)为0.916~6.875,平均值为2.078。多态性信息含量(PIC)为0.136~0.771,平均值为0.457。有2个SSR位点的Ho/He值大于1,说明其杂合度较高。对元宝枫9个主要分布区的250个无性系进行遗传多样性分析,结果表明:种源间存在着11%的遗传变异,同源内无性系之间具有85%的遗传变异。聚类分析和主坐标分析结果一致,9个主要分布区的元宝枫聚为3大类群:赤峰和泰安两个分布区的亲缘关系较近,聚为一大类群;北京、泗水、淄博、滕州、海阳和蒙山亲缘关系次近,聚为一类;四川的单独聚为一类。本研究结果为元宝枫种质资源保存和利用提供了理论支持。In order to study the genetic diversity of Acer truncatum germplasm resources,the DNA of Acer truncatum clonal populations was amplified by fluorescent PCR,and the PCR products were detected by capillary electrophoresis to analyze the genetic differences and genetic diversity among concentrated distribution regions and homologous clones of Acer truncatum.11 pairs of fluorescent labeled SSR primers were used to analyze 250 Acer truncatum clones and 102 alleles were detected.The number of alleles at each locus ranged from 4 to 16,with the average of 9.27.The observed heterozygosity(Ho) was 0.42~0.94,and was 0.66 on average.The expected heterozygosity(He) was 0.45~0.88,and the average expected heterozygosity was 0.73.Shannon's diversity index(I)was 0.916~6.875,and the average value was 2.078.The polymorphism information content(PIC) ranged from 0.136to 0.771,with an average value of 0.457.The Ho/He ratios of two SSR loci were greater than 1,which indicated the high heterozygosity of the two SSR loci.Genetic diversity of 250 clones from 9 main distribution areas of Acer truncatum was analyzed.The results showed that there were 11% genetic variation among provenances and 85%genetic variation among congeners.The results of cluster analysis and principal coordinate analysis were consistent,9 main distribution areas of Acer truncatum bunge were clustered into 3 groups:Chifeng and Tai'an distribution areas were close and clustered into one group;Beijing,Surabaya,Zibo,Tengzhou,Haiyang and Mengshan were close and clustered into one group;Sichuan was isolated into one group.These results provide theoretical supports for the conservation and utilization of Acer truncatum bunge germplasm resources.
