摘 要:为了从连翘中提取出高效低毒的抗肿瘤活性物质,试验采用水煮提取、乙醇沉淀、大孔吸附树脂柱层析、薄层层析的方法从连翘中分离提取到连翘抗肿瘤活性物质FS-4,通过超高效液相色谱仪串联四级杆飞行时间质谱仪鉴定FS-4的化学成分,并研究了FS-4对SGC-7901的增殖抑制作用及超微结构的影响。结果表明:从连翘中分离提取到14种化学成分,分别由咖啡酸、齐墩果酸、白桦脂酸、熊果酸、软脂酸、硬脂酸6种已知成分和8种未知成分组成。FS-4对SGC-7901的增殖具有抑制作用,FS-4浓度越高,时间越长对其增殖抑制作用越大,具有剂量时间依赖性;在6,12,24,36小时时,FS-4作用于SGC-7901的半数抑制浓度(IC50)分别为87.38,56.10,34.45,3.23μg/mL。FS-4作用SGC-7901后,其核浆比明显减小,核仁消失,胞浆中出现明显的空泡,染色质密集边缘化,细胞表面绒毛脱失,且质膜呈现叶片样突起,并有凋亡小体形成。说明从连翘中分离、提取得到的FS-4具有抗肿瘤活性,可抑制SGC-7901的增殖及诱导其凋亡。In order to extract antitumor active substances with high efficiency and low toxicity from Forsythia suspensa,in this experiment,the antitumor active substance(FS-4)of Forsythia suspensa was isolated and extracted from Forsythia suspensa by boiled water extraction,ethanol precipitation,macroporous adsorption resin column chromatography and thin layer chromatography.FS-4 was identified by ultra-high performance liquid chromatograph tandem quadrupole time-of-flight mass spectrometer.The effect of FS-4 on the proliferation inhibition and ultrastructure of SGC-7901 was also studied.The results showed that 14 chemical components were isolated and extracted from forsythia,which were composed of 6 known components of caffeic acid,oleanolic acid,betulic acid,ursolic acid,palmitic acid and stearic acid,and 8 unknown components.FS-4 had inhibitory effect on the proliferation of SGC-7901;the higher the concentration of FS-4 and the longer time it lasted,the greater the inhibitory effect on proliferation;and it was dose-time-dependent.At 6,12,24,and 36 h,the half inhibitory concentrations(IC50)of FS-4 acting on SGC-7901 were 87.38,56.10,34.45,and 3.23μg/mL,respectively.After FS-4 action SGC-7901,its nucleusplasma ratio decreased significantly;nucleoli disappeared;obvious vacuoles appeared in the cytoplasm;chromatin was densely marginalized;cell surface villi were lost,and the plasma membrane showed leaf-like protrusions;and the formation of apoptotic bodies confirmed that FS-4 had the effect of inducing apoptosis.The results suggested that FS-4 isolated and extracted from Forsythia suspensa had antitumor activity,which could inhibit the proliferation of SGC-7901 and induce its apoptosis.
