益气养阴活血方对IgA肾病小鼠外周血B淋巴细胞C1β-3-GaLT11、Cosmc表达及肠黏膜免疫反应的影响

摘  要:目的:探究益气养阴活血方对IgA肾病小鼠外周血B淋巴细胞核心1β1,3-半乳糖基转移酶(C1β-3GaLT11)及其伴侣蛋白(Cosmc)表达和肠道黏膜免疫反应的影响。方法:小鼠随机分为空白组、模型组、中药(益气养阴活血方)低、中、高剂量组,每组15只。除空白组外,其余各组建立小鼠IgA肾病模型。检测血清白蛋白(Alb)、尿素氮(BUN)、血肌酐(Scr)水平;检测血清Gd-IgA1水平及肾小球系膜区Gd-IgA1荧光强度;HE染色及PAS染色观察肾组织病理改变;HE染色观察结肠组织病理变化;qRT-PCR法检测外周血B淋巴细胞Cosmc mRNA、C1β-3-GaLT11 mRNA表达;Western Blot法检测结肠组织中Cosmc、C1β-3-GaLT11、B细胞活化因子(Baff)、TGF-β表达及肾组织中肿瘤坏死因子α(TNF-α)、白细胞介素17(IL-17)、TGF-β蛋白表达水平。结果:与空白组相比,模型组小鼠Gd-IgA1沉积、蛋白尿和血尿症状增加,肾功能降低,肾小球系膜增生及肾小管炎症浸润明显,肠黏膜炎症浸润等病理损伤严重,肠道及外周血B淋巴细胞表面的Cosmc及C1β-3-GaLT11活性降低,Gd-IgA1生成增多(P<0.05)。益气养阴活血方可降低Gd-IgA1沉积,改善IgA肾病小鼠蛋白尿和血尿症状,促进肾功能恢复,缓解肾小球系膜增生及肾小管炎症浸润,缓解肠黏膜炎症浸润,提高肠道及外周血B淋巴细胞表面的Cosmc、C1β-3-GaLT11活性,抑制Gd-IgA1生成(P<0.05)。结论:益气养阴活血方可改善肠黏膜损伤、提高肠道及外周血B淋巴细胞表面的Cosmc、C1β-3-GaLT11活性,使其催化IgA糖基化能力增强,减少Gd-IgA1生成及沉积,缓解Gd-IgA1诱发的IgA肾病肾损伤。Objective:To explore the effects of Yiqi Yangyin Huoxue Recipe on the expression of core 1β1,3-galactosyltransferase(C1β-3GaLT11)and its chaperone protein(Cosmc)in peripheral blood B lymphocytes and intestinal mucosal immune response in mice with IgA nephropathy.Methods:Mice were randomly divided into blank group,model group,and low,medium and high dose groups of Chinese medicine(Yiqi Yangyin Huoxue Recipe),with 15 mice in each group.Except for the blank group,the other groups established mouse IgA nephropathy models.the levels of serum albumin(Alb),urea nitrogen(BUN),and blood creatinine(Scr);The level of serum gd-iga1 and the fluorescence intensity of gd-iga1 in glomerular mesangial area were detected;HE staining and PAS staining were used to observe the pathological changes of kidney tissue;HE staining was used to observe the pathological changes of colon tissue;qRT-PCR was used to detect the expression of Cosmc mRNA and C1β-3-GaLT11 mRNA in peripheral blood B lymphocytes;Western Blot method was used to detect the expression levels of Cosmc,C1β-3-GaLT11 and B cell activating factor(Baff)in colon tissue and the levels of tumor necrosis factor-α(TNF-α),interleukin-17(IL-17)and TGF-βproteins in kidney tissue.Results:Compared with the blank group,the model group mice had increased Gd-IgA1 deposition,increased proteinuria and hematuria,decreased renal function,obvious glomerular mesangial hyperplasia and renal tubule inflammation infiltration,and serious pathological damage such as intestinal mucosal inflammatory infiltration,the activities of Cosmc and C1β-3-GaLT11 on the surface of B lymphocytes in the intestine and peripheral blood decreased,and the production of Gd-IgA1 increased(P<0.05).Yiqi Yangyin Huoxue Recipe could reduce Gd-IgA1 deposition,improve proteinuria and hematuria in IgA nephropathy mice,promote renal function recovery,relieve glomerular mesangial hyperplasia and renal tubular inflammation infiltration,relieve intestinal mucosal inflammatory infiltration,and increase the activities of Cos
