C/GP/Co水凝胶联合肌腱干细胞在大鼠跟腱损伤愈合中的作用研究

摘  要:目的探讨用壳聚糖/β-甘油磷酸/胶原(chitosan/β-glycerophosphate/collagen,C/GP/Co)水凝胶包裹肌腱干细胞(tendon stem cells,TSCs)注射至大鼠跟腱损伤部位对大鼠急性跟腱损伤的愈合作用。方法选取96只SD大鼠制作跟腱缺损模型,随机分为4组,每组24只,分别为:对照组、水凝胶组、TSCs组、TSCs联合水凝胶治疗组。每组大鼠分别于损伤后2、4、6周采用二氧化碳(CO_(2))麻醉法各处死8只,用组织学方法检测各组I型胶原面积,实时荧光定量分析各组成肌腱相关基因表达水平,Western blot方法检测各组成肌腱分化相关蛋白表达水平,评估损伤跟腱的愈合情况。结果与对照组、水凝胶组及TSCs组相比,TSCs联合水凝胶治疗组明显促进了大鼠跟腱损伤的愈合。在损伤4周及6周时,TSCs联合水凝胶治疗组的I型胶原面积明显大于其他3组,Ⅰ型胶原及成腱分化相关基因Scleraxis(SCX)mRNA的表达量、Ⅰ型胶原及SCX蛋白表达水平均明显优于其他3组(P均<0.05)。但TSCs联合水凝胶治疗组的Ⅲ型胶原面积占比、Ⅲ型胶原mRNA的表达量及蛋白表达水平明显低于其他3组(P<0.05)。结论C/GP/Co水凝胶联合TSCs治疗肌腱损伤不仅促进了肌腱损伤愈合的速度,也提高了其愈合质量。Objective To investigate the healing effect of chitosan/β-glycerophosphate/collagen(C/GP/Co)hydrogel filled with tendon stem cells(TSCs)on acute Achilles tendon injuries in a rat model.Methods The model of Achilles tendon defect was made in 96 SD rats,which were randomly divided into 4 groups:control group,hydrogel group,TSCs group and TSCs hydrogel group(n=24 for each).At 2,4 and 6 weeks after injury,8 rats in each group were respectively sacrificed by CO_(2) anesthesia.For all of them,the area of type I collagen was measured by using the histological method,and the expression levels of tendon-related genes were analyzed by using real-time quantitative PCR and tendon differentiation-related proteins by western blot to evaluate the healing of the injured Achilles tendon.Results Compared with the other three groups,the TSCs+hydrogel group revealed much quicker healing of the Achilles tendon injury.At 4 and 6 weeks of injury,it showed a significantly larger area of type I collagen,and much higher mRNA expressions and protein expressions of type I collagen and Scleraxi,compared with the other three groups respectively(all P<0.05).Moreover,the area of type III collagen and the mRNA and protein expression of type III collagen expressions in TSCs+hydrogel group were significantly lower than those in the other three groups,indicating less scar formation.Conclusion TSCs+hydrogel not only quickens the healing speed but also promotes the healing quality following rat Achilles tendon injury.This study provides a theoretical and experimental basis for further related research.
