摘  要:目的为深入探索变应性鼻炎(Allergic Rhinitis,AR)的发病机制,从微生物学角度出发,比较AR患者与健康人的菌群组成差异,微生物标记物及代谢产物。方法通过收集40名AR患者和20名健康人的收集鼻腔分泌物,利用16SrDNA技术在V3和V4区域测序及KEGG通路富集分析,以鉴别AR患者的鼻腔菌群标记物和代谢产物。结果微生物多样性没有显著差异,在门水平上3种菌门差异明显,纲水平上有5种,目、科、种、属均有差异,其中纲水平上:AR组a-变形菌(3%)、梭状芽胞杆菌(17%)、放线菌(21%)等高于健康组,杆菌(22%)、厚壁菌门(2%)低于健康组,关键物种有a-变形菌(P=0.034)、莫拉氏菌科(P=0.015),不动杆菌属(P=0.006),假单孢杆菌目(P=0.01),伯克霍尔德氏菌目(P=0.027)和约翰逊不动杆菌(P=0.004),可能作为AR的微生物标记物。KEGG富集分析中AR组与健康组,两组间没有显著差异(P>0.05),但两组中有相同的显著差异代谢产物(P<0.05)。结论这些显著差异的菌群有可能作为诊断AR的微生物标记物,为今后临床诊断和治疗提供参考。Objective In order to explore the pathogenesis of allergic rhinitis(AR),we compared the differences in microbiota composition between AR patients and healthy individuals,and the microbial markers of AR from a microbiological point of view.Methods Nasal secretions were collected from 40 AR patients and 28 healthy individuals,sequenced in the V3 and V4 regions using 16SrDNA technology and KEGG pathway enrichment analysis to identify nasal bacterial markers and metabolites in AR patients.Results The nasal diversity of healthy people was slightly higher than that of AR patients,and there were significant differences in the three phyla at the phylum level,with five species at the class level,and the orders,families,species,and genera were different,at the class level:A-proteus(3%),Clostridium(17%),Actinomyces(21%),etc.in AR group were higher than those in healthy group,while Bacillus(22%)and Firmicute(2%)were lower than those in healthy group,the key species were A-Proteus(P=0.034),Moraxella(P=0.015),Acinetobacter(P=0.006).Pseudomonas(P=0.01),Burkholderella(P=0.027)and Acinetobacter Johnson(P=0.004)may be microbial markers for AR.In KEGG enrichment analysis,there was no significant difference between the AR group and the healthy group(P>0.05),but there were the same significant metabolites differences between the two groups(P<0.05).Conclusion These significantly different flora may be used as microbial markers for the diagnosis of AR and provide a reference for future clinical diagnosis and treatment.
