摘 要:目的探讨半乳糖凝集素-1(Galectin-1)预处理对机械通气相关性肺损伤(VILI)小鼠的影响。方法选择清洁级健康雄性C57BL/6小鼠30只,6~8周龄,体重22~30 g。采用随机数字表法将小鼠分为三组:对照组(C组)、VILI组(V组)和Galectin-1+VILI组(G组),每组10只。C组气管插管后保持自主呼吸4 h,V组和G组气管插管后机械通气4 h。气管插管前1 h C组和V组腹腔注射生理盐水0.75 ml,G组腹腔注射Galectin-13μg。于气管插管前即刻、自主呼吸或机械通气结束时采集动脉血检测PaO_(2),后处死小鼠,收集肺泡支气管灌洗液(BALF),采用ELISA法检测BALF中IL-1β和IL-18浓度。取肺组织测定湿/干重比(W/D),采用qRT-PCR法检测肺组织GSDMD、caspase-1和caspase-11 mRNA表达量,Western blot法检测肺组织GSDMD、caspase-1和caspase-11蛋白含量,HE染色法观察病理改变并行肺损伤评分。结果与C组比较,V组和G组机械通气结束时PaO_(2)明显降低,W/D、BALF中IL-1β和IL-18浓度、肺组织GSDMD、caspase-1、caspase-11 mRNA表达量及蛋白含量、肺损伤评分明显升高(P<0.05)。与V组比较,G组机械通气结束时PaO_(2)明显升高,W/D、BALF中IL-1β和IL-18浓度、肺组织GSDMD、caspase-1、caspase-11 mRNA表达量及蛋白含量、肺损伤评分明显降低(P<0.05)。结论Galectin-1能够升高小鼠PaO_(2),降低IL-1β和IL-18浓度、经典及非经典细胞焦亡途径相关基因mRNA表达量和蛋白含量,减轻小鼠VILI。Objective To investigate the effect of galectin-1 preconditioning on pyroptosis of ventilator-induced lung injury(VILI)in mice.Methods Thirty clean grade healthy male C57BL/6 mice,aged 6-8 weeks,weighing 22-30 g,were divided into three groups by random number table method:control group(group C),VILI group(group V),and galectin-1+VILI group(group G),10 mice in each group.After endotracheal intubation,group C kept spontaneous breathing for 4 hours,groups V and G kept mechanical ventilation for 4 hours.One hour before endotracheal intubation,groups C and V were intraperitoneally injected with normal saline 0.75 ml,and group G was intraperitoneally injected with galectin-13μg.Arterial blood was collected before endotracheal intubation and after spontaneous respiration or ventilation to detect PaO_(2).Then mice were sacrificed and bronchoalveolar lavage fluid(BALF)was collected.Concentrations of IL-1βand IL-18 in BALF were detected by ELISA.Lung tissue was collected for determination of the wet weight/dry weight ratio(W/D).The expression of GSDMD,caspase-1,and caspase-11 mRNA and protein in lung tissues were detected by qRT-PCR and Western blot.Pathological changes of the lungs were observed and scored by HE staining.Results Compared with group C,PaO_(2) were significantly decreased,W/D,concentrations of IL-1βand IL-18 in BALF,mRNA and protein expressions of GSDMD,caspase-1 and caspase-11,and lung injury score were significantly increased in groups V and G(P<0.05).Compared with group V,PaO_(2) was significantly increased,W/D,concentrations of IL-1βand IL-18 in BALF,mRNA and protein expressions of GSDMD,caspase-1,and caspase-11,and lung injury score were significantly decreased in group G(P<0.05).Conclusion Galectin-1 can increase PaO_(2) in mice and reduce IL-1βand IL-18 concentration,mRNA expression and protein content of classical non-classical pyroptosis pathway related genes,and reduce VILI in mice.
