摘 要:为了鉴定天津市某养殖场鲤鱼腹腔内寄生的“面条样”虫体,本试验通过PCR方法扩增虫体的18S rRNA、COⅠ和COB基因序列,并进行分子鉴定和基因测序,分析3种基因的同源性、分子进化和系统发育关系。结果显示,寄生于鲤鱼体内的虫体的18S rRNA、COⅠ和COB基因序列分别与双线绦虫分离株MN204040.1(中国)、MN219466.1(中国)和EU241209.1(法国)的核苷酸序列同源性较高,为99.45%、98.74%和99.26%。基于18S rRNA、COⅠ和COB基因序列构建的系统发育树显示,本试验分离的绦虫裂头蚴与GenBank数据库中已鉴定的双线绦虫聚在同一分支上,且与双线绦虫中国武汉分离株的种属亲缘关系较近。本试验初步阐明获得的天津市双线绦虫分离株与其他种属之间的系统发育关系,为双线绦虫的分子鉴定和系统发育关系分析提供了数据支持。To identify the"noodle-like"worm parasites found in the abdominal cavities of common carp from a fish farm in Tianjin,PCR was used to amplify the 18S rRNA,COⅠ,and COB gene sequences of the worms.Molecular identification and gene sequencing were performed,and the homology,molecular evolution,and phylogenetic relationships based on these three genes were analyzed.The results indicated that the 18S rRNA,COⅠ,and COB gene sequences of the worms parasitizing the Cyprinus carpio showed high homology with nucleotide sequences from Digramma interuota isolates MN204040.1(China),MN219466.1(China),and EU241209.1(France),with homologies of 99.45%,98.74%,and 99.26%,respectively.Phylogenetic trees constructed based on the 18S rRNA,COⅠ,and COB gene sequences showed that the tapeworm plerocercoid isolated in this study clustered in the same branch as the Digramma interuota species identified in the GenBank database,and they were closely related to the Digramma interuota isolate from Wuhan,China.This study provides initial insights into the phylogenetic relationship of the Digramma interuota isolate obtained in Tianjin with other species,supporting molecular identification and phylogenetic analysis of Digramma interuota.
