摘 要:为了解鸡胚来源大肠杆菌的致病性和耐药性情况,本试验对山东省某白羽肉鸡孵化场6个不同批次种蛋孵化后随机抽检的174枚啄壳未出壳死胚的卵黄和胎粪进行了大肠杆菌分离。通过细菌培养、形态观察、生化试验方法鉴定大肠杆菌;通过小鼠攻毒试验和毒力基因PCR检测确定大肠杆菌分离株的致病性;利用纸片扩散法药敏试验检测大肠杆菌分离株的耐药性。结果显示,共分离鉴定大肠杆菌28株;小鼠攻毒试验结果显示,27大肠杆菌分离株对小鼠的致死率达到了50.0%以上,其中有24株超过了83.0%;在检测的14种毒力基因中,papA、papC、eaeA和vat四种基因未检出,fimC检出率最高(85.7%),另外9种毒力基因检出率介于3.6%~64.3%;菌株的致病性与携带的毒力基因数量大致呈正相关;耐药性检测结果显示,大肠杆菌分离株对10种抗菌药物具有不同程度的敏感性,耐药率介于17.9%~89.3%。结果表明,本试验分离的鸡胚源大肠杆菌绝大多数具有致病性,对临床常见抗菌药物存在不同程度的耐药。本试验结果为种鸡场加强大肠杆菌垂直传播的管理和控制以及临床用药的选择提供了参考依据。In order to understand the pathogenicity and drug resistance of Escherichia coli(E.coli)from chicken embryos,this study isolated E.coli from the yolk and embryonic feces of 174 randomly sampled unhatched dead embryos from six different batches of eggs in a broiler hatchery in Shandong Province.The strains were isolated and identified using bacterial culture,morphological observation,and biochemical tests.The pathogenicity of the isolated E.coli strains was determined through mouse inoculation tests and PCR detection of virulence genes.The drug resistance of the isolates was assessed by the paper disk diffusion method.The results showed that 28 strains of E.coli were isolated and identified.In the mouse inoculation test,27 E.coli isolates had a mortality rate over 50.0%,with 24 strains exceeding 83.0%.Among the 14 tested virulence genes,papA,papC,eaeA,and vat were not detected,while fimC had the highest detection rate(85.7%),and the detection rates of the other nine virulence genes ranged from 3.6%to 64.3%.There was a positive correlation between the pathogenicity of the strains and the number of carried virulence genes.Drug resistance testing showed that E.coli isolates had varying degrees of sensitivity to 10 antibiotics,with resistance rates ranging from 17.9%to 89.3%.The results indicate that majority of the isolated E.coli from chicken embryos are pathogenic,showing different degrees of resistance to commonly used clinical antibiotics.This study provides reference information for the management and control of vertical transmission of E.coli in breeder farms and the selection of antibiotics in clinical practice.
