摘  要:以辣椒育种资源9-68为研究对象,利用cDNA-AFLP技术对辣椒果实不同发育期进行分析,并进行生物信息学分析,探索辣椒果实发育中相关基因的差异表达,以期获得辣椒果实发育的相关基因及表达信息。结果表明:利用42对引物组合共扩增出1 926条TDF(transcriptic derived fregment),其中包括上调表达、下调表达、无差异表达等类型。选择特异表达的25个TDF进行回收测序,获得19个清晰的序列结果。其中8个TDF序列在Blastn库中检索到相似性较高及功能确定的mRNA、cDNA或基因序列,8个TDF序列与Blastx库中已登录的mRNA或cDNA序列相似性较高,但是具体功能未确定。另外3个TDF在GenBank、EST库的比对分析中,没有搜索到与3个TDF有较高相似性的结果,可能为首次发现的碱基序列。Taking pepper breeding resources 9-68 as the research object,cDNA-AFLP technology was used to analyze different developmental stages of pepper fruits,and bioinformatics analysis was conducted to explore the differential expression of related genes during pepper fruit development,in order to obtain relevant genes and expression information for pepper fruit development.The results showed that a total of 1926 TDFs(transcriptive derived fragments)were amplified using 42 primer combinations,including up-regulated expression,down-regulated expression,and undifferentiated expression types.Selecting 25 specifically expressed TDFs for recovery sequencing,19 clear sequence results were obtained.Among them,8 TDFs sequences were found in the Blastn library to have known cDNA,mRNA,or gene sequences with high similarity,while 8 TDFs sequences were found in the Blastx library to have cDNA or mRNA sequences with high similarity that had been registered in the database,but their specific functions had not been determined.In the comparison analysis of three TDFs in GeneBank and EST libraries,no results with high similarity were found,which may be the first discovered base sequence.
