朱红膏对中性粒细胞分泌炎症因子及凋亡的影响

摘　　要：目的观察朱红膏对中性粒细胞(PMN)炎症因子分泌和凋亡的影响,研究其促进慢性皮肤溃疡愈合的机制。方法原子荧光光谱仪检测汞的含量,磁珠法提取PMN,CCK-8法检测细胞活性;CBA法检测IL-1β、IL-8的含量;Annexin V-PI法检测PMN凋亡;Real Time-PCR法检测PMN Caspase-3 mRNA和Caspase-9 mRNA的表达;Western blot法检测Cytochrome C、Bcl-2和Bax蛋白表达。结果选择汞含量0.96、0.48、0.24 mg/L分别作为朱红膏的高、中、低三个浓度组;与模型组相比,高浓度组显著性抑制PMN分泌IL-1β(P<0.01),各组均显著性抑制的IL-8分泌(P<0.01),促进PMN凋亡(P<0.01)、Cytochrome C和Bax蛋白表达(P<0.01),降低Bcl-2蛋白表达(P<0.01)。结论朱红膏通过抑制中性粒细胞分泌炎症因子和促进凋亡治疗慢性皮肤溃疡。Objective To observe the Effect of Zhu Hong Ointment(ZHO)on secretion of inflammatory factors and apoptosis of neutrophils,and study its mechanism of promoting the healing of chronic skin ulcer.Methods The content of Hg was detected by atomic fluorescence spectrometer,PMN was extracted by magnetic bead method,and cell activity was detected by CCK-8 meth-od;detection the content of IL-1βand IL-8 by CBA.PMN apoptosis was detected by Annexin V-PI method.The expression of Caspase-3 mRNA and Caspase-9 mRNA in PMN was detected by Real Time-PCR.Western blot was used to detect the ex-pression of PMN Cytochrome C,Bcl-2 and Bax protein.Results The Hg content of 0.96 mg/L,0.48 mg/L and 0.24 mg/L were selected as the high,medium and low concentration groups of ZHO.Compared with the model group,the high concentration group significantly inhibited the secretion of IL-1β(P<0.01).All groups significantly inhibited the secretion of IL-8 in PMN(P<0.01),promoted the apoptosis of PMN(P<0.01),the expression of Cytochrome C and Bax protein(P<0.01),and decreased the expression of Bcl-2 protein(P<0.01).Conclusion Zhu Hong Ointment can promoted the healing of chronic skin ulcers by inhibiting the secretion of inflammatory factors and promoting apoptosis of neutrophils.
