基于PI3K/Akt信号通路探讨茯苓杏仁甘草汤抗动脉粥样硬化巨噬细胞凋亡的作用机制

摘　　要：目的探讨茯苓杏仁甘草汤对内毒素(lipopolysaccharide,LPS)联合氧化低密度脂蛋白(oxidized low density lipopro-tein,ox-LDL)诱导巨噬细胞凋亡的干预作用。方法构建巨噬细胞凋亡模型,检测巨噬细胞凋亡率、细胞Bax与Bcl-2 mRNA及蛋白表达水平、PI3K、Akt蛋白磷酸化水平,检测茯苓杏仁甘草汤对上述指标的影响,及PI3K抑制剂LY294002干预后上述指标的变化。结果茯苓杏仁甘草汤降低Bax与Bcl-2 mRNA及蛋白表达水平比值,升高PI3K、Akt蛋白磷酸化水平,其作用受到LY294002抑制。结论茯苓杏仁甘草汤抑制巨噬细胞凋亡,其机制与激活PI3K/Akt信号通路有关,为临床治疗巨噬细胞凋亡相关心血管疾病提供了实验依据。Objective To study the effects of Fuling Xingren Gancao Formula on macrophages apoptosis induced by lipopolysac-charide(LPS)and oxidized low-density lipoprotein(ox-LDL).Methods The model of macrophages apoptosis was construc-ted.The macrophages apoptosis rate of macrophages,the mRNA and protein expression levels of Bax and Bcl-2,the phosphoryl-ation levels of PI3K and Akt protein,the effects of Fuling Xingren Gancao Formula on the above indexes were detected,and the changes of the above indexes were detected after adding PI3K inhibitor LY294002.Results Fuling Xingren Gancao Formula de-creased the ratio of Bax to bcl-2 mRNA and protein expression,increased the level of phosphorylation PI3K and Akt protein,and its effects were partially inhibited by LY294002.Conclusion Fuling Xingren Gancao Formula inhibits macrophages apopto-sis,and its mechanism is related to the activation of PI3K/Akt signal pathway,which provides an experimental basis for the clinical treatment of macrophage apoptosis related cardiovascular diseases.
