摘 要:目的探讨柿蒂鞣质对D-型半乳糖诱导的衰老小鼠体内抗氧化、抗炎能力的影响。方法48只C57小鼠随机分为6组即正常对照组、衰老模型组、维生素E(Vit E)阳性对照组、柿蒂鞣质高、中、低剂量给药组。小鼠通过颈皮下注射D-型半乳糖建立衰老模型,阳性对照组与柿蒂鞣质治疗组每天接受药物灌胃进行干预。8周后,检测小鼠血清及肝脏、心脏和脾脏中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)的活性和丙二醛(MDA)含量;测定血清中抗肿瘤坏死因子(TNF-α)、白细胞介素-6(IL-6)及白细胞介素1β(IL-1β)含量水平。结果柿蒂鞣质可抑制小鼠血清和肝脏、心脏、脾脏中MDA的生成(P<0.01),提高SOD与GSH-Px等活力表达(P<0.01);有效降低血清中TNF-α、IL-6及IL-1β等抗炎因子含量(P<0.01)。结论柿蒂鞣质能有效提高衰老小鼠的抗氧化和抗炎能力,其主要作用机理可能与抑制脂质过氧化的生成、增强抗氧化酶活力表达及减轻机体抗炎因子水平等有关。Objective To study the effects of tannins from Persimmon calyx.on antioxidant and anti-inflammation in mouse aging model induced by D-galactose.Methods Forty-eight mice were randomly divided into six groups:the normal control group,aging model group,vitamin E positive control group,and tannins from Persimmon calyx.groups respectively at high,medium and low dose.The activities of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px),and malondialdehyde(MDA)content were determined in serum,liver,spleen and heart,and antitumor necrosis factor(TNF-α),interleukin 6(IL-6)and interleukin 1 beta(IL-1β)were determined in serum after 8-week intragastric administration.Results Tannins from Persim-mon calyx.improved the activities of SOD and GSH-Px,decreased the content of MDA,and increased the levels of TNF-α,IL-6 and IL-1β.Conclusion Tannins from Persimmon calyx.can enhance antioxidant and anti-inflammatory capacity of mouse aging model induced by D-galactose.
