摘 要:目的:选取国内6家制造商的尼龙及聚醚砜针头过滤器开展洗脱及吸附试验,研究针头过滤器在色谱试验中的洗脱及吸附效应。方法:洗脱试验选取有机试剂(甲醇、乙腈),使用尼龙针头过滤器过滤,观察液相色谱洗脱峰情况;吸附试验配置不同浓度林可霉素、克拉霉素与罗红霉素标准品溶液,使用针头过滤器过滤,LC-MS测定,计算回收率。结果:6家制造商的尼龙针头过滤器均有可被乙腈与甲醇洗脱的残留物峰;不同制造商聚醚砜针头过滤器吸附效应不同,同一制造商的聚醚砜针头过滤器对不同化合物的吸附程度也存在差异。被过滤溶液的浓度变小,需要弃去的初滤液体积有变多趋势。随着弃去的初滤液体积变多,被吸附的物质存在被释放的可能。结论:提示试验人员使用针头过滤器进行色谱前处理时应充分评估针头过滤器的洗脱及吸附效应。Objective:To study the elution and adsorption effect of nylon and polyether sulfone needle filters in chromatographic tests from 6 domestic manufacturers.Methods:Organic reagents(methanol and acetonitrile)were selected for elution test and filtered by nylon needle filters.The elution peaks were detected by liquid chromatography.Standard solutions of lincomycin,clarithromycin and roxithromycin with different concentrations for adsorption test were filtered by needle filters and determined by LC-MS,and the recovery rate was calculated.Results:There were residue peaks which could be eluted by acetonitrile and methanol in nylon needle filters from 6 manufacturers.The adsorption effect of polyether sulfone needle filters from different manufacturers was different,and the adsorption degree of polyether sulfone needle filters from the same manufacturer was also different.As the concentration of filtered solution decreases,the volume of initial filtrate to be discarded tends to increase.As the volume of discarded filtrate increases,the adsorbed material was likely to be released.Conclusion:It is suggested that the elution and adsorption effects of needle filters should be fully evaluated when using needle filter for chromatographic pretreatment.
